Overexpression of Human Apolipoprotein A-II in Mice Induces Hypertriglyceridemia Due to Defective Very Low Density Lipoprotein Hydrolysis

Abstract: Two lines of transgenic mice, hAIItg-␦ and hAIItg-, expressing human apolipoprotein (apo)A-II at 2 and 4 times the normal concentration, respectively, displayed on standard chow postprandial chylomicronemia, large quantities of very low density lipoprotein (VLDL) and low density lipoprotein (LDL) but greatly reduced high density lipoprotein (HDL). Hypertriglyceridemia may result from increased VLDL production, decreased VLDL catabolism, or both. Post-Triton VLDL production was comparable in transgenic and cont… Show more

“…Given the prior evidence for a role of APOA2 in insulin sensitivity, HDL concentrations, and NEFA concentrations [6,21,41,42,43], we examined the effect of APOA2 variation and haplotypes on ten related traits: NEFA, HDL cholesterol, triglycerides, fasting insulin, fasting glucose, 60 and 120 min glucose during an OGTT and a measure of insulin secretion at 60 min that is normalized for the insulin sensitivity (OCI60; see Methods). Surprisingly, we found little evidence for an effect of either individual variants or haplotypes on triglycerides, HDL cholesterol, NEFA, or fasting insulin.…”

“…The up-regulation of human APOA5 is important regarding of the molecular mechanism of triglycerides homeostasis action of fibrate. Indeed, APOA5 represents to date the first example of an apolipoprotein whose overexpression leads to a decrease in triglyceride levels (9,20), whereas APOC3 or another apolipoprotein transgene leads to an increase in triglycerides plasma levels (21)(22)(23)(24).…”

“…Plasma very low density lipoprotein (VLDL)/ chylomicron-TG clearance in mice is completely inhibited under these conditions. Blood samples were taken from each mouse at 0, 30, 90, and 150 min after injection (26). The accumulation of newly synthesized VLDL/chylomicron-TG was measured in plasma aliquots and ascertained by FPLC.…”
