Overproduction of valuable methoxylated flavones in induced tetraploid plants of Dracocephalum kotschyi Boiss

Abstract: Background: Ploidy manipulation is considered an efficient method to increase production potential of medicinally important compounds. Dracocephalum kotschyi Boiss. is an endangered medicinal plant of Iran. Various concentrations of colchicine (0.05, 0.10, 0.20, and 0.50% w/v) were applied to shoot apical meristems of D. kotschyi seedlings in two and four-leaf stages to induce tetraploidy.

“…Similar results were reported in cannabis by Bagheri and Mansouri (2015). By contrast, a higher number of trichomes have been reported for some polyploids (Aina et al, 2012;Zahedi et al, 2014). A relationship between the number of trichomes in leaves and resistance to abiotic stresses has also been reported (Huang et al, 2007) in Dioscorea zingiberensis (Zhang et al, 2010b) and Coccira palmate (Ntuli and Zobolo, 2008).…”

“…Phenolic compounds are active in pathogen resistance, pigment production, and protection against ultraviolet radiation (Winkel-Shirley, 2001); they also qualitatively and quantitatively improve the biochemical profile of plants (Maruška et al, 2010). Polyploidy often enhances the expression and concentration of some secondary metabolites and defence chemicals (DeJesus-Gonzalez and Weathers, 2003;Jones et al, 2008;Nilanthi et al, 2009;Dhooghe et al, 2011;Dehghan et al, 2012;Hsia et al, 2013;Gomes et al, 2014;Zahedi et al, 2014).…”

“…Reports showed high methoxylated flavonoid content in the Labiatae family. The xanthomicrol concentration in the aerial parts and leaves of D. Kotschyi has been reported to be about 1.384 mg g −1 dry wt . The rosmarinic acid content in hairy root lines of D. Kotschyi ranged from 10 to 1500 mu g g −1 DW, with a peak of 15 times higher than that in intact control roots.…”

“…Thus, three methods are necessary for screening and establishing pure polyploid strains. Polyploid shows the importance of genetic diversity and plant evolution (Zahedi et al 2014).…”
