Overproduction of Wild-Type and Bioengineered Derivatives of the Lantibiotic Lacticin 3147

Abstract: Lacticin 3147 is a broad-spectrum two-peptide lantibiotic whose genetic determinants are located on two divergent operons on the lactococcal plasmid pMRC01. Here we introduce each of 14 subclones, containing different combinations of lacticin 3147 genes, into MG1363 (pMRC01) and determine that a number of them can facilitate overproduction of the lantibiotic. Based on these studies it is apparent that while the provision of additional copies of genes encoding the biosynthetic/production machinery and the regul… Show more

“…It has been reported that mutagenized LtnA1 and LtnA2 are each still modified, processed, and transported and retain antimicrobial activity (7)(8)(9). In all cases, the mutagenized substrates were still highly homologous to their natural counterparts.…”

“…In Bacillus subtilis ATCC 6633, additional copies of subtilin self-protection (immunity) genes spaIFEG have been integrated into the genome of the producer strain which enhanced the subtilin tolerance level and concomitantly, the subtilin yield [8]. Furthermore, the bioengineered derivatives of the lantibiotic lacticin 3147 were overproduced when additional copies of genes encoding the biosynthetic/production machinery and the regulator ltnR were present [5]. However, the enhancement of the bacteriocin productions has been identified in the overnight cultures in all of the studies mentioned above.…”

“…The main ones being the producer bacterial family, their molecular weight and finally their amino acid sequence homologies and/or gene cluster organization [59,70]. Based on a relatively recent approach [69,71,72] bacteriocins produced by LAB have been categorized into two major classes: the lanthionine -containing bacteriocins or lantibiotics (class I) and the largely unmodified linear peptide antimicrobials (class II).…”
