Overview of Electrochemical DNA Biosensors: New Approaches to Detect the Expression of Life

Abstract: DNA microarrays are an important tool with a variety of applications in gene expression studies, genotyping, pharmacogenomics, pathogen classification, drug discovery, sequencing and molecular diagnostics. They are having a strong impact in medical diagnostics for cancer, toxicology and infectious disease applications. A series of papers have been published describing DNA biochips as alternative to conventional microarray platforms to facilitate and ameliorate the signal readout. In this review, we will consid… Show more

“…Figure reproduced with permission of the publisher from ref. 27 Due to the favourable hybridisation properties of PNAs (higher thermal duplex stability and mismatch discrimination), DNA biosensors with PNA recognition layers have been developed. 28 PNA-derived biosensors are able to distinguish closely related sequences under less restrictive reaction conditions than for DNA.…”

“…Amplified DNA targets can be analyzed for the presence of SNP by different direct and indirect hybridization strategies including intercalator dyes in allele-specific digital PCR (dPCR) [17], sequence-specific fluorogenic probes [18], electrochemical sensors [19][20][21][22][23][24] and diagnostic nano-materials [25]. Electrochemical sensing is a promising approach for SNP detection which takes advantage of synthetic oligonucleotide probes and the large thermodynamic changes in enthalpy and entropy.…”

“…nano-particles and fluorescence spectroscopy [17][18][19][20][21][22][23][24][25]. Next several efficient colorimetric assays for SNP sensing in amplified DNA have been developed based on modified gold nano-particles (Au-nps; Figure 2) [26].…”

“…DNA is one of the most important targets that have been studied during the last decades. Applications range from medical diagnosis and genome sequencing to food, pollution, and environmental analysis [6]. There are several methods to detect DNA: the most widely used is typically based on microarray technique, which consists on nucleic acid hybridization and optical readout [35].…”
