RESUMO.O objetivo deste estudo foi avaliar o potencial ovulatório de diferentes doses de prostaglandina E2 em camundongos fêmeas pré-púberes. Foram utilizadas 18 fêmeas BALB/c pré-púberes, tratadas com 5 UI de eCG via intraperitoneal (i.p.), no Dia 0. No Dia 2, os animais foram divididos aleatoriamente em 6 grupos, para receberem via i.p.; 1) 0.5 mL de PBS (n = 3), 2) 5 µg de GnRH (n = 3), 3) 25 µg de PGE2 (n = 3), 4) 50 µg de PGE2 (n = 3), 5) 150 µg de PGE2 (n = 3) e 6) 250 µg de PGE2 (n = 3). No Dia 3, as fêmeas foram mortas por deslocamento da coluna cervical e os ovidutos foram coletados para contagem de oócitos através da técnica de transiluminação sob estereomicroscópio. Não houve diferença no número de camundongos fêmeas que ovularam e o número de oócitos ovulados nas fêmeas tratadas com GnRH e 250 µg de PGE2, e ambos os grupos apresentaram diferenças em relação ao grupo PBS (P < 0.001). Os resultados deste estudo demonstram que a PGE2 na dose de 250 µg é capaz de induzir a ovulação em camundongos fêmeas pré-púberes. The objective of this study was to evaluate the effect of different doses of prostaglandin E2 (PGE2) on the ovulation of prepubertal female mice. Eighteen In BALB/c prepubertal mice were treated with 5 IU of intraperitoneal (i.p.) eCG on day 0. On day 2, the mice were randomly distributed into 6 groups for the i.p. administration of: 1) 0.5 mL of PBS (n = 3), 2) 5 µg of GnRH (n = 3), 3) 25 µg of PGE2 (n = 3), 4) 50 µg of PGE2 (n = 3), 5) 150 µg of PGE2 (n = 3) e 6) 250 µg of PGE2 (n=3). On day 3, the mice were killed and the oviducts were collected for the oocyte count by transillumination technique under stereomicroscopy. There were no difference in the number of ovulated mice and number of ovulated oocyte in the females treated with GnRH and 250 µg de PGE2, and both groups showed differences in comparison with PBS group (P<0.001). The results demonstrated that the PGE2 dose of 250 µg induces the ovulation in prepubertal female mice.

Since structural luteolysis involves deterioration of tissue, the gene expression of matrix-metalloproteinase-1 (MMP-1) and the respective tissue inhibitor of this metalloprotease (TIMP-1) were measured at various times on the day of pro-oestrus and in animals in which the preovulatory prolactin surge was blocked for the duration of 3 cycles by bromocriptine. An additional group of prolactin-blocked rats received a prolactin replacement injection on the afternoon of pro-oestrus. In spontaneously pro-oestrous rats, MMP-1 and TIMP-1 gene expression increased significantly (P < 0.01) prior to the occurrence of the preovulatory LH surge but simultaneously with the onset of the preovulatory prolactin surge. When prolactin release was blocked by bromocriptine for 3 cycles, no such changes were observed during the afternoon of pro-oestrus. However, an intraperitoneal injection of bovine prolactin at the time when the preovulatory prolactin surge occurs normally, increased MMP-1 and TIMP-1 gene expression (P < 0.01). These results indicate that MMP-1 and TIMP-1 gene expression are stimulated by the preovulatory prolactin surge. Previous work has shown that the preovulatory LH surge activates the enzymatic cascade which leads to increased collagenase activity.
