Oxidative Inactivation of Liver Mitochondria in High Fructose Diet-Induced Metabolic Syndrome in Rats: Effect of Glycyrrhizin Treatment

Abstract: Metabolic syndrome is a serious health problem in the present world. Glycyrrhizin, a triterpenoid saponin of licorice (Glycyrrhiza glabra) root, has been reported to ameliorate the primary complications and hepatocellular damage in rats with the syndrome. In this study, we have explored metabolic syndrome-induced changes in liver mitochondrial function and effect of glycyrrhizin against the changes. Metabolic syndrome was induced in rats by high fructose (60%) diet for 6 weeks. The rats were then treated with … Show more

“…The present study demonstrated for the first time that treatment with GLY significantly reduced serum total and LDL cholesterol levels without affecting HDL cholesterol levels and that GLY reduced atherosclerotic lesion sizes in the aortic sinuses of Apoe -/- mice on HFD. The effects of GLY on lipid metabolic dysregulation are mediated by several mechanisms, including inhibition of circulatory stress hormones, augmentation of muscular lipid uptake, inhibition of mitochondrial oxidative stress and aconitase degradation, and up-regulated expression of the activator of PXR, the nuclear pregnane X receptor, which can also affect the energy metabolism through changes in fatty acid metabolism [13-15]. We propose that GLY plays an important role in ameliorating lipid metabolism and AS.…”

“…Glutathione plays an essential role in maintaining intracellular redox state ( Asensi et al, 1999 ). Decreases in GSH level and GSH/GSSG ratio in liver and brain have been reported after prolonged high-fructose supplementation ( Song et al, 2012 ; Jarukamjorn et al, 2016 ; Sil and Chakraborti, 2016 ). A similar decrease in GSH/GSSG ratio of skeletal muscle after long-term fructose drinking in this study indicates that the observed effect on redox state is systemic.…”

“…Rats subjected to this diet presented high levels of ROS, reduced glutathione content, and reduced aconitase activity. The administration of glycyrrhizin can attenuate these alterations [185].…”
