Oxidative Stimulation of Glutathione Synthesis in Arabidopsis thaliana Suspension Cultures

Abstract: A system based on Arabidopsis thaliana suspension cultures was established for the analysis of glutathione (CSH) synthesis in the presence of hydrogen peroxide. Mild oxidative stress was induced by use of the catalase inhibitor, aminotriazole, and its development was monitored by measurement of the oxidative inactivation of aconitase. Addition of 2 mM aminotriazole resulted in a 25% decrease in activity of aconitase over 4 h. During the subsequent 10 h, no further decrease in aconitase activity was measured de… Show more

“…Furthermore, GSH plays a protective role in salt tolerance by maintaining the redox state (Shalata et al 2001). The increased level of the GSH pool is generally regarded as a protective response against oxidative stress (May and Leaver 1993;Xiang and Oliver 1998), although defense against stress situations sometimes occurs irrespective of the GSH concentration (Potters et al 2004). In this study, Cd stress caused a significant increase in GSH content (Fig.…”

“…Plant Material-The derivation and maintenance of cultures of Arabidopsis (12), tobacco (13), tomato (14), and French bean (15) have been described previously. The carrot cultures (previously unpublished) were grown in Murashige and Skoog (16) basal salts supplemented with 3% sucrose and 2 mg/ml 2,4-D.…”

“…To deplete glutathione levels, we grew Arabidopsis seedlings on Murashige and Skoog (1962) medium, supplemented with a nontoxic and highly specific inhibitor of the first enzyme of GSH synthesis, L-buthionine-sulfoximine (BSO; Griffith and Meister, 1979;May and Leaver, 1993). Inclusion of BSO in the growth medium resulted in a strong reduction in SA-induced glutathione levels (data not shown).…”
