Oxidative Stress and Antioxidant Status in Mouse Liver: Effects of Dietary Lipid, Vitamin E and Iron ,

Abstract: The purpose of this study was to determine the effects of dietary fat, vitamin E and iron on oxidative damage and antioxidant status. Male Swiss-Webster mice (1 mo old) were fed a basal vitamin E-deficient diet that contained either 8% fish oil + 2% corn oil or 10% lard with or without 1 g dl-alpha-tocopheryl acetate. The diets without vitamin E contained either 0.21 or 0.95 g ferric citrate/kg. Diets were fed for 4 wk/kg diet. Compared with the vitamin E-supplemented groups, mice fed diets without vitamin E (… Show more

“…Since IL-4 is known to suppress the production of other reactive nitrogen and oxygen species in addition to NO (32)(33)(34)(35)(36), and these intermediates may also be involved in mediating liver pathology (37)(38)(39), the generation of O 2 Ϫ , hydrogen peroxide, and ONOO Ϫ by granuloma cells was investigated. To examine O 2 Ϫ production, granuloma cells were stimulated with PMA, and O 2 Ϫ was detected by measuring the formation of blue formazan after addition of NBT.…”

“…eicosapentaenoic acid (EPA), 20 : 5(n-3) and docosahexaenoic acid (DHA), 22 : 6(n-3)) of fish oils are extremely susceptible to lipid peroxidation, even in the presence of added dietary antioxidants (Gonzalez et al, 1992). The data evaluating the effect of fish oil n-3 PUFA on oxidative stress and lipid peroxidation markers in vivo vary with both animal and clinical studies reporting increased oxidation (Ibrahim et al, 1997), no effect on oxidation (Nenseter et al, 1992) and even some indication of decreased susceptibility to induced oxidation (Yuan and Kitts, 2002).…”

“…Fish are the major source of EPA ϩ DHA, but fish oil supplements produce a decreased oxidative stability of plasma LDL (15,16). In contrast, consumption of fish itself may be protective against type 2 diabetes (17).…”
