Oxymatrine inhibited cell proliferation by inducing apoptosis in human lung cancer A549 cells

Abstract: Abstract. To investigate the inhibition effect of oxymatrine induces human lung cancer A549 cells apoptosis. The A549 cells were cultured for 24 h, than the various concentration of oxymatrine (2 mmol/L, 4 mmol/L, 8 mmol/L, 15 mmol/L) were added into different experimental group cells, and 5-fluorouracil were added into the positive control group cells for 12 h, 24 h, 36 h, 48 h respectively. The A549 cells inhibition rate, apoptosis, and the expression of Bcl-2 and Bax were examined by MTT method, Annexin /PI … Show more

“…OMT has been extensively investigated for antitumor effects against various cancers [15], including lung [12], gastric [13], pancreatic [11], and breast [14]. However, to the best of our knowledge, the antitumor effects of OMT on GBM have yet to be fully investigated.…”

“…It is widely understood that caspase-3 is an essential terminal caspase that executes apoptosis and eventually leads to DNA fragmentation in the mitochondrial pathway [19, 21]. Therefore, induction of apoptosis may represent a potential approach for antitumor therapy [22], and there is an increasing body of evidence that OMT may induce apoptosis in a number of cancer cell lines [11, 12, 15, 18, 2326]. The data presented herein adds to this body of evidence by demonstrating that OMT induces apoptosis, decreases expression of Bcl-2, and concurrently increases expression of Bax and caspase-3.…”

“…It has been demonstrated that oxymatrine promotes anti-tumor properties in the setting of many carcinomas, including colon carcinoma, hepatocellular carcinoma and lung cancer. [19][20][21][22] However, the signaling pathways which mediate its anti-tumor properties are divergent. In colorectal cancer, epithelial-mesenchymal transition (EMT), which is involved in the initiation of metastasis for malignant cancer progression was inhibited by oxymatrine through regulation of the nuclear factor-kappaB (NF-κB) signaling pathway.…”

“…It is also known that a mixture of oxymatrine and micellar nanoparticles is an effective proliferation inhibitor of SMM7721 cells [52]. Oxymatrine treatment significantly induces apoptosis by increasing Bax protein expression and reducing Bcl-2 in human lung cancer A549 cells [53]. Proteomic analysis has shown that oxymatrine induces apoptosis in HeLa cells by inhibiting inosine monophosphate dehydrogenase type II (IMPDH2), mitochondrial related apoptotic protein [54].…”
