P2X3 Receptor Involvement in Pain States

Abstract: The understanding of how pain is processed at each stage in the peripheral and central nervous system is the precondition to develop new therapies for the selective treatment of pain. In the periphery, ATP can be released from various cells as a consequence of tissue injury or visceral distension and may stimulate the local nociceptors. The highly selective distribution of P2X(3) and P2X(2/3) receptors within the nociceptive system has inspired a variety of approaches to elucidate the potential role of ATP as … Show more

“…6 Immunostaining for P2X 1 (a, e), P2X 2 (b, f) the most to extracellular nucleotide activation, suggesting a predominately nociceptive role for P2X receptors [28]. P2X 3 receptor, in particular, has been found to be selectively expressed on small-diameter capsaicin-positive nociceptive neurons [29], and homomeric P2X 3 and heteromeric P2X 2/3 receptors have roles in acute and longer lasting sensitivity to nociception, respectively [30].…”

“…A review includes a discussion of the role of P2X3 receptors in inflammatory pain [102]. During the inflammatory process in peripheral tissue, neither prostaglandins nor sympathetic amines can sensitize primary afferent neurons by themselves; they depend on previous neuronal P2X3 receptor activation [103].…”

“…neuropathic and inflammatory pain, headache pain and migraine) [22,[31][32][33][34][35][36][37][38][39]. As a consequence, targeting these receptors represents an innovative therapeutic strategy to treat both neuropathic and inflammatory pain conditions [40,41]. Recent papers described the rapid transition of the P2X3 receptors to a desensitization state followed by a relatively slow recovery of sensitivity to ATP [13,42].…”
