P2Y1R and P2Y2R: potential molecular triggers in muscle regeneration

Abstract: Muscle regeneration is indispensable for skeletal muscle health and daily life when injury, muscular disease, and aging occur. Among the muscle regeneration, muscle stem cells’ (MuSCs) activation, proliferation, and differentiation play a key role in muscle regeneration. Purines bind to its specific receptors during muscle development, which transmit environmental stimuli and play a crucial role of modulator of muscle regeneration. Evidences proved P2R expression during development and regeneration of skeletal… Show more

“…In general, the high-water permeability of cell membranes causes rapid osmotic responses via the movement of osmotically active ions into or out of cells. Hypertrophy of the cell volume is observed when signaling from P2Y2 is transduced in muscles [ 35 ].…”

“…We show that cellular stress (as induced by pro‐inflammatory LPS) causes an increase in CTKs associated with the tRNA degradation pathway. The increased presence of CTKs in response to LPS is particularly relevant given the importance of purinergic signaling in skeletal muscle (Fulle, 2015 ; Wang et al., 2023 ) and the putative role for tRNA‐derived fragments in regulating skeletal muscle homeostasis and stress responses (Liapi et al., 2020 ). Collectively, iP forms were the most abundant of all CTKs detected both within and outside the cell (Figure 4 ) and all forms of iP (FB, RB, and NT) were elevated in response to LPS treatment in the extracellular space (chemical structures of iP as a FB, RB, or NT are summarized in Figure 5a ).…”

“…The action of P2Y receptors is controversial. P2Y1 and P2Y2 receptors improve muscle regeneration, which is blocked when these receptors are inhibited [112]. However, in skeletal muscle fibroblasts, P2Y2 receptors promote fibrosis and muscle atrophy via Akt/ERK/PKC activation [113].…”
