Pyrin domain (PYD), a subclass of protein motif known as the death fold, is frequently involved in inflammation and immune responses. PYD modulates nuclear factor-kappa B (NF-κB) signalling pathway upon various stimuli. Herein, a novel recombinant pyrin domain protein (RPYD) was generated. Its role and mechanism in inflammatory response in an ovalbumin (OVA) induced asthma model was investigated. After OVA challenge, there was inflammatory cell infiltration in the lung, as well as airway hyper-responsiveness (AHR) to inhaled methacholine. In addition, eosinophils increased in the bronchoalveolar lavage fluids, alone with the elevated levels of Th-2 type cytokines [interleukin (IL)-4, IL-5 and IL-13], eotaxin, and adhesion molecules. However, the transnasal administration of RPYD before the OVA challenge significantly inhibited these asthmatic reactions. Moreover, RPYD markedly suppressed NF-κB translocation, reduced phosphorylation of p38 MAPK, and thus attenuated the expression of intercellular adhesion molecule 1 and IL-6 in the BEAS-2B cells stimulated by proinflammatory cytokines in vitro. These findings indicate that RPYD can protect asthma host from OVAinduced airway inflammation and AHR via down-regulation of NF-κB and p38 MAPK activities. The accessory gland (AG) is an important component of the male reproductive system of arthropods, its secretions enhance fertility, some AG proteins bind to the spermatozoa and affect its function and properties. Here we report the first comprehensive catalog of the AG secreted fluid during the mature phase of the Chinese mitten crab (Eriocheir sinensis). AG proteins were separated by one-dimensional gel electrophoresis and analyzed by reverse phase high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS). Altogether, the mass spectra of 1173 peptides were detected (1067 without decoy and contaminants) which allowed for the identification of 486 different proteins annotated upon the NCBI database (http://www.ncbi.nlm.nih.gov/) and our transcritptome dataset. The mass spectrometry proteomics data have been deposited at the ProteomeXchange with identifier PXD000700. An extensive description of the AG proteome will help provide the basis for a better understanding of a number of reproductive mechanisms, including potentially spermatophore breakdown, dynamic functional and morphological changes in sperm cells and sperm acrosin enzyme vitality. Thus, the comprehensive catalog of proteins presented here can serve as a valuable reference for future studies of sperm maturation and regulatory mechanisms involved in crustacean reproduction.

The c-Jun N-terminal protein kinase mitogen-activated protein kinases (JNK MAPKs) are an evolutionarily conserved family of serine/threonine protein kinases involved in controlling cell growth, differentiation, and apoptosis in mammalian testis and germ cells. As a scaffold protein of the JNK pathway, JNK-interacting protein 4 (JIP4) is found in some mammalian spermatozoa and participates in certain male reproductive processes. Few have reported on the roles of JNK and JIP4 in the Chinese mitten crab Eriocheir sinensis H. Milne Edwards, 1853. Based on transcriptome sequences from the testis of E. sinensis, we successfully cloned full-length cDNAs of JNK and JIP4 (designated Es-JNK and Es-JIP4, respectively). Western blotting showed relatively high expression of Es-JNK and Es-JIP4 proteins in the testis. Expression levels of total Es-JNK, phosphorylated Es-JNK (Es-p-JNK), and Es-JIP4 proteins decreased gradually during spermatogenesis. Es-JNK and Es-JIP4 mRNAs were mainly localized in the nucleus from the spermatogonium stage to the secondary spermatocyte stage and were widely distributed in the nuclear cup, apical cap, acrosomal tubule, and acrosomal vesicle of spermatids. Es-JNK, Es-p-JNK, and Es-JIP4 proteins were nevertheless mainly dispersed in the cytoplasm and intercellular substance of spermatogonium and primary spermatocytes, in the nucleus of secondary spermatocytes, and in the apical cap, acrosomal tubule, acrosomal vesicle, and nuclear cup of spermatids. During the acrosome reaction induced by the calcium ionophore A23187, Es-JNK, Es-p-JNK, and Es-JIP4 proteins were found mainly expressed in the apical cap and acrosomal vesicle by immunofluorescence microscopy. The results implicate functions for Es-JNK and Es-JIP4 in spermatogenesis and the acrosome reaction in E. sinensis.
