p53 and breast cancer, an update

Abstract: Abstractp53 plays a key role in mediating cell response to various stresses, mainly by inducing or repressing a number of genes involved in cell cycle arrest, senescence, apoptosis, DNA repair, and angiogenesis. According to this important function, p53 activity is controlled in a very complex manner, including several auto-regulatory loops, through the intervention of dozens of modulator proteins (the 'p53 interactome'). p53 mutations are observed in a significant minority of breast tumours. In the remaining … Show more

“…To characterize the molecular differences between these two groups of cell lines, we first listed the cell lines' molecular status and subtypes (Table 1; refs. [15][16][17][18]. A previous study suggested that NVP-BEZ235 induces cell death in breast cancer cells with PIK3CA mutations or HER2 amplification, whereas all of the PTEN deleted, mutated, or silenced cell lines are insensitive to NVP-BEZ235 (5).…”

“…Lactoferrin is found to bind with insulin-like growth factor-binding protein-3 [97], thereby act by altering the availability or stability of insulin-like growth factor, a known regulator of the phosphatidylinositol 3-kinase/Akt pathway [98]. Accordingly, camel urine possess supreme anti-proliferative effect on many lines of cancer cell possibly through the induction of the cyclin dependent kinase inhibitor p21 as it is found to up-regulates p21 in the p53-defective cancer cells [39,99]. Anti-proliferative effect of camel urine is significantly influenced by breeds, age, sex and other camel characteristics.…”

“…Physiological Significance of p53 Activation by MYBBP1A-p53 plays a critical role in cancer prevention, given that p53 suppresses tumorigenesis by inducing cell cycle arrest and apoptosis through its transcriptional activity (71). We recently demonstrated that MYBBP1A plays a role in tumor prevention in the context of p53 activation (72).…”
