Prolactinomas are the most prevalent functional pituitary adenomas that cause chronic pathological hyperprolactinemia. Prolactin is known to promote cell growth and inhibit apoptosis in cells. Paeoniflorin is the principal component of radix Paeoniae alba (the main ingredient in some traditional herbal formulas clinically used for hyperprolactinemia-associated disorders). Recent findings from intensive studies have suggested that paeoniflorin regulates cell proliferation and apoptosis in many cell lines. However, the effects of paeoniflorin in pituitary tumor cells remain unknown. Here the results by the Cell Counting Kit-8 and colony formation assays showed that paeoniflorin concentration-dependently decreased cell viability in both MMQ and GH3 cells and colony formation in GH3 cells, suggesting inhibition of cell proliferation by paeoniflorin. By flow cytometry, paeoniflorin was found to increase apoptotic rate in MMQ cells. Mechanistically, Western blot results revealed that paeoniflorin enhanced protein expression of cleave caspase-9 and -3, and Bax, whereas it suppressed Bcl-2 protein expression in MMQ cells. Furthermore, paeoniflorin upregulated phosphorylated p53 protein expression, but it decreased prolactin concentration and prolactin protein expression in both MMQ and GH3 cells. Thus, the present results demonstrate that paeoniflorin inhibits cell proliferation and induces the mitochondrial pathway-mediated apoptosis in prolactinoma cells. These antitumor property is associated with inhibition of prolactin secretion. Our findings may provide new insight into the mechanisms underlying improving prolactinoma-associated disorders of paeoniflorin-enriched herbs and formulas.

Glycyrrhiza uralensis Fisch., known as licorice, is one of the most famous traditional Chinese medicines. In this study, we perform a metabolome analysis using liquid chromatography-tandem mass spectrometry to assign bioactive components in different parts of licorice from different geographical origins in Gansu province of China. Sixteen potential biomarkers of taproots from different geographical origins were annotated, such as glycycoumarin, gancaonin Z, licoricone, and dihydroxy kanzonol H mainly exist in the sample of Jiuquan; neoliquiritin, 6 ′ -acetylliquiritin, licochalcone B, isolicoflavonol, glycyrol, and methylated uralenin mainly exist in Glycyrrhiza uralensis from Lanzhou; gancaonin L, uralenin, and glycybridin I mainly exist in licorice from Wuwei for the first time. Substance abuse is one of the problems that many countries are currently dealing with and it imposes several complications on our communities. The dopamine reward system is one of the regions of the brain that plays important roles in the pleasure and persistence of addiction. This review was conducted to investigate the effects and action mechanisms of medicinal plants on the dopaminergic review system in preventing substance abuse. The key words "dopamine*" in combination with "substance abuse", "drug abuse", "addiction" or "medicinal plant", "herb*", and "phyto*" were used to retrieve relevant publications indexed in the Institute for Scientific Information (ISI) and PubMed using the EndNote software. After examining the studies, we included 23 of them in final analysis. Medicinal plants and their derivatives can exhibit anti-addiction effects in substance use disorders mainly through influencing dopamine receptors (D1 and D2). They can also serve as appropriate alternatives to drugs and to help treat relapse due to stimulating the dopamine reward system. Because substance abuse is multifactorial, it should be taken into account that phytotherapy alone cannot be effective in treating it but instead a combination of different therapies should be adopted to fight it.
