Landscape features (e.g., mountains and rivers) can act as barriers to dispersal and gene flow, and therefore impede population connectivity, increasing genetic differentiation between populations. The concave‐eared torrent frog (Odorrana tormota) is a rare, stream‐associated species in eastern China. In this study, we investigated the genetic structure and population demography of this narrowly distributed frog based on mitochondrial Cyt b gene and seven nuclear microsatellite loci. As a result, we found that the rare frog still preserved a relatively high level of genetic diversity compared to some other amphibia. Population structure analyses distinctly identified three or four tentative genetic clusters for the species in the study area. Additionally, by fine‐scale spatial autocorrelation analysis, significant positive genetic structure was uncovered in the shortest distance classes (0–5 km, 5–10 km). Demographic analyses revealed a population expansion (0.075–0.017 Mya) and 15 times population decline (c. 9000 years ago). In conclusion, we supposed that stable montane environments and associated historical population expansion might provide an opportunity for the species to harbor high genetic diversity. The relatively recent population decline might be correlated with climate change as well as genetic differentiation among populations. In addition, our results showed that, on a small landscape scale, dispersal is closely linked to geographic distance and the presence of river systems may not substantially affect the genetic structure for the narrowly distributed O. tormota.

Diversification patterns and demography of montane species are affected by Pleistocene climate fluctuations. Empirical cases from the Qinling Mountains (QM) region, which is a major biogeographic divider of East Asia, are few. We used DNA sequence data of the complete mitochondrial ND2 gene to detect effects of the Pleistocene glaciations on phylogeographic profiles of a frog species, Feirana taihangnica, which is endemic to the QM. Four distinct lineages consisting of seven sublineages were revealed. The strongest signal of biogeographical structure (F ct = 0.971, P \ 0.01) was found when populations were grouped according to these seven sublineages. One narrow secondary contact zone was detected in the middle QM between the lineage from middle QM and the lineage from eastern QM. Coalescent simulations indicated that this species colonized the QM region by a stepping-stone model. Divergences among lineages had likely been influenced by the uplift of the Tibetan Plateau during the late Miocene-tolate Pleistocene, as well as by the Pleistocene climatic cycles. Coalescent simulations also suggested that F. taihangnica populations have persisted through the Pleistocene glacial periods in multiple refugia across the QM region. Demographic analyses indicated that all lineages, except the lineage in the Funiu Mountains, have been experienced postglacial expansion of population size and distribution range. In conclusion, Pleistocene climate fluctuations and tectonic changes during the late Miocene-late Pleistocene have profoundly influenced the phylogeography and historical demography of F. taihangnica.

Aim The extent to which the ranges of temperate biota in East Asia have been shaped by geological and climatic changes during the Pleistocene remains poorly understood. Previous molecular-based studies of the greater horseshoe bat (Rhinolophus ferrumequinum), which is widespread across China and Japan, suggest that this species was able to persist in multiple refugia in this region. However, limited availability of samples precluded a detailed analysis of past distributions. By integrating ecological niche modelling with phylogenetic analyses, we aim to resolve the impact of past events on the population genetic structure of R. ferrumequinum and provide detailed descriptions of potential distribution patterns during this time.Location Central/East China and Japan.Methods Phylogenetic analyses were carried out using 1098 bp of the mitochondrial ND2 gene and 13 microsatellite loci from bats sampled at 26 localities across China and Japan. Haplotype data were used in isolation-withmigration models (IMa) to estimate divergence dates between different lineages, and mismatch distribution analyses were undertaken to test for signs of population expansion. Geographical structuring of the microsatellite data was examined by spatial clustering. Ecological niche modelling was performed based on highresolution palaeoclimate data inferred for the Last Glacial Maximum (LGM).Results Phylogenetic analyses identified deep levels of divergence between three major lineages broadly corresponding to Central China, East China and Japan. The divergence of all three lineages was estimated to have occurred before the LGM. A strong correlation was observed between the ecological niche model and phylogenetic data, with both methods inferring that R. ferrumequinum populations in Central China did not undergo significant range contractions during glacial episodes. The impact of glacial cycles on East China would have been more severe, with some locations being unable to support viable populations. Zones of secondary contact were identified between Central/East China and East China/Japan, which probably result from some historical gene flow between isolated populations.Main conclusions Integrating ecological niche modelling within phylogeographical studies provides support for a complex demographic history for R. ferrumequinum in East Asia, as well as persistence during glacial episodes in multiple refugia across their range.
