Palindromic sequence impedes sequencing-by-ligation mechanism

Abstract: BackgroundCurrent next-generation sequencing (NGS) platforms adopt two types of sequencing mechanisms: by synthesis or by ligation. The former is employed by 454 and Solexa systems, while the latter by SOLiD system.
