Palmitic acid is a toll-like receptor 4 ligand that induces human dendritic cell secretion of IL-1β

Abstract: Palmitic acid (PA) and other saturated fatty acids are known to stimulate pro-inflammatory responses in human immune cells via Toll-like receptor 4 (TLR4). However, the molecular mechanism responsible for fatty acid stimulation of TLR4 remains unknown. Here, we demonstrate that PA functions as a ligand for TLR4 on human monocyte derived dendritic cells (MoDCs). Hydrophobicity protein modeling indicated PA can associate with the hydrophobic binding pocket of TLR4 adaptor protein MD-2. Isothermal titration calor… Show more

“…Cytokines lead to NASH development through the activation of the nuclear factor κB (NF‐κB) and C‐Jun NH 2 ‐terminal kinase (JNK) pathway. FAs, particularly saturated FA, were found to activate the toll‐like receptor 4 (TLR4), activating both the NF‐κB and JNK pathway . In the liver, TLR4 is expressed by hepatocytes, Kupffer cells and stellate cells.…”

“…FAs, particularly saturated FA, were found to activate the toll-like receptor 4 (TLR4), activating both the NF-κB and JNK pathway. 21,22 In the liver, TLR4 is expressed by hepatocytes, Kupffer cells and stellate cells. Upon phagocytosis of cholesterol, Kupffer cells are activated and TLR4 expression is upregulated.…”

“…4-гексадеканова (пальмітинова кислота) -найпоширеніша у природі жирна кислота, входить до складу гліцеридів більшості тваринних жирів, рослинних олій і воску. Відомо, що вона стимулює протизапальні реакції в імунних клітинах людини через TLR4-рецептори [8], їй притаманна протипухлинна й антидіабетична активності [9].…”

“…In addition, palmitate can act as a TLR4 ligand on dendritic cells and induce IL-1B secretion. 63 This may be specified that palmitate is a natural compound becoming a candidate for tuberculosis drug. CYP2B6 is one of cytochrome P450 enzyme involved in the transformation of drug and other xenobiotics, CYP2B6 polymorphism can be an indicator for tuberculosis treatment.…”
