Paracrine signaling in stem cell renewal and in neoplastic tumor growth

Abstract: Paracrine pathway activities are being increasingly recognized as instrumental regulatory mechanisms of epithelial-stromal interactions that play important roles in physiological and pathological self-renewal of stem cells and in the initiation and maintenance of neoplastic tumor development. Stromal-specific Hedgehog (Hh) responses and epithelial-associated Wnt pathway activities have been recently appreciated as important factors in stem cell self-renewal and carcinogenesis. Furthermore, Hh and Wnt pathways … Show more

“…Transforming growth factor  (TGF-), Wnt and hedgehog signaling are pivotal pathways that influence the cell division, invasion, migration and ulcer repair processes Lagasse, 2008;Zhao, 2014). Disorders in these processes usually lead to gastric adenocarcinoma and other tissue-specific gastrointestinal cancers (Stojnev et al, 2014).…”

“…The important distinction of this focus is the long and interesting list of cellular features that all adult stem cells have in common, not on the complex spectrum of distinctive phenotypes that the stem cells can become. The unexpected and simplifying conclusion is that all adult stem cells have a similar profound paracrine activity that is essential for the normal turnover and regeneration especially active and discernable at sites of injury [1][2][3][4][5][6].…”

“…Zhao Chen [4], in his article, discussed paracrine signaling activities in stem cell renewal and neoplastic tumor growth. The interaction between epithelial cells and stromal cells is key to embryonic development, adult tissue homeostasis and tumorigenesis.…”
