Paraspeckle Protein NONO Promotes TAZ Phase Separation in the Nucleus to Drive the Oncogenic Transcriptional Program

Abstract: The Hippo pathway effector TAZ promotes cellular growth, survival, and stemness through regulating gene transcription. Recent studies suggest that TAZ liquid-liquid phase separation (LLPS) compartmentalizes key cofactors to activate transcription. However, how TAZ LLPS is achieved remains unknown. Here, it is shown that the paraspeckle protein NONO is required for TAZ LLPS and activation in the nucleus. NONO is a TAZ-binding protein. Their interaction shows temporal regulation parallel to the interaction betwe… Show more

“…Whilst a role for NONO-driven phase separation in paraspeckle formation is yet to be established, NONO phase separation is required for radiation-induced DNA damage repair (Fan et al, 2021). In addition, NONO also promotes phase separation and activation of TAZ, the hippo pathway effector, to drive oncogenic transcription and tumorigenesis in glioblastoma, although a direct role for phase separation of NONO itself was not addressed in that study (Wei, Luo et al, 2021). Thus, NONO phase separation may be important for many of its regulatory activities in different contexts.…”

“…Overexpression of NONO promoted nuclear TAZ phase separation, while low expression of NONO decreased nuclear TAZ phase separation. Moreover, the low expression of NONO inhibited the interaction of TAZ with enhancers and TEAD [147]. In addition, the phase separation of LATS1 can be promoted by phosphatidic acid-binding lncRNA SNHG9, thereby promoting carcinogenic YAP signaling.…”

“…NONO, an RNA/DNA binding protein, drives oncogenic transcriptional programs to promote tumorigenesis. 33 NONO interacts with the hypoxia inducible factor-1/2 complex and its downstream transcripts to activate the transcription of hypoxia-induced genes, which facilitate HCC progression. 34 CENPI, a member of the centromere protein family, has an essential role in accurate chromosome segregation.…”
