Abstract:Among the 17 members of Poly (ADP-ribose) polymerase enzyme family, PARP-1 is a very crucial DNA repair enzyme and the level of PARP-1 is found elevated in several cancers such as ovarian, triple negative breast cancer. Inhibitors of PARP -1 is a new feature of anticancer drug development. In this mini, we covered the mode of action, recent development and approval of PARP-1 inhibitors. We also provide an insight on designing strategy of novel PARP-1 inhibitors and future direction.
