Particle and Gel Characterization of Irinotecan-Loaded Double-Reverse Thermosensitive Hydrogel

Abstract: The irinotecan-loaded double-reverse thermosensitive hydrogel (DRTH) is a dispersed system of irinotecan-loaded solid lipid nanoparticles (SLN) in a thermosensitive hydrogel. To optimise the particle and gel properties of DRTHs for rectal administration of irinotecan, SLNs and DRTHs were prepared with tricaprin, triethanolamine, Tween 80, and Span 20. Among the SLNs tested, an SLN composed of 1 g irinotecan, 0.5 g lipid mixture, and 0.5 g combined surfactant gave the highest entrapment efficiency and smallest … Show more

“…Then, 0.5 g ALP-loaded NLCs gel was applied on the lower (upright glass slide) and it was adjusted, using adjustable pan, such that both the glass vials gently attached each other. Then, weights were placed in an ascending order on the other side of the balance, until the detachment of the two slides occur [ 39 , 40 ]. The weight that was required to detach the two glass vials was noted as the bio-adhesive strength.…”

“…The ability of a semisolid formulation to spread on the skin plays a vital role in the administration of a standard medicated formulation and thus in the efficacy of a topical therapy [80]. It is reported earlier and demonstrated in this study that there is a direct correlation between spreadability of a vesicle-loaded pharmaceutical gel and its in vivo therapeutic efficacy [81]. The higher values of spreadability indicate the ease of application which leads to an increase in the surface area for drug application and penetration [82].…”
