Passage Number of 4T1 Cells Influences the Development of Tumour and the Progression of Metastasis in 4T1 Orthotopic Mice

Abstract: Background: The study was aimed to elucidate the influence of passage number of 4T1 cells for the development of the ideal tumour model. Methods: A total of 24 female BALB/c mice was divided equally into three groups: i) control (phosphate buffered saline [PBS] only); ii) group A (subjected to 4T1 cells of passage number 9) and iii) group B (subjected to 4T1 cells of passage number 10). The injections were introduced at the 3rd mammary pad of the mice. The net volume of the tumours was examined. Histopatholog… Show more

“…29 Additional attempts to make experiments more repeatable would also involve ensuring that tumor sizes are similar between experiments at the time of randomization 30 and potentially use cell line samples which have undergone a sufficiently similar amount of passages. 31 The effect of RT is relatively consistent between studies, which explains why the RSE% for parameters associated with APC recruitment and T cell exhaustion are highly precise and why the VPCs capture the RT monotherapy effects well. Taken together, these findings justify the large cohort sizes in each experiment, which gives extensive data points for more improved fitting with less residual error and better internal validation.…”

“…Additional data, such as cage cleaning cycles, should also be recorded, as the microbiome is also known to affect the tumor response to various drugs 29 . “…To determine the effect of the cell culture on protein expression, the same experiment was repeated across five subcultures of the reference cell line obtained from ATCC. These subcultures are known as “passages” [42] . It is important to allow the cells to fully recover after being thawed from liquid nitrogen storage.…”

“…Cells were passaged in a 70%–80% confluence within 2–3 days of post‐culture and were harvested by 1× trypsin EDTA (DNA Biotech Co, Tehran, Iran). 7 × 10 5 4T1 cells suspended in 100 μL PBS were injected subcutaneously into the right flank of each mouse 25,26 . After 12 days, the tumor appeared as expected on the right side.…”
