Passage number of cancer cell lines: Importance, intricacies, and way‐forward

Abstract: Cancer cell lines play a crucial role as invaluable models in cancer research, facilitating the examination of cancer progression as well as the advancement of diagnostics and treatments. While they may not perfectly replicate the original tumor, they generally exhibit similar characteristics. Low-passage cancer cell lines are generally preferred due to their closer resemblance to the original tumor, as long-term culturing can alter the genetic and molecular profiles of a cell line thereby highlighting the imp… Show more

“…The cell culture was incubated under standard culture conditions at 37 °C with 5% CO 2 in a humidified atmosphere and sub-cultured every 2 to 3 days. To ensure the maintained cell phenotype, the cells were used for experiments up to the 10th passage [ 24 ]. All cell lines were regularly controlled for mycoplasma contamination using a MycoProbe mycoplasm detection kit (R & D Systems, Inc., Minneapolis, MN, USA).…”

“…Researchers must carefully select the appropriate passage number to maintain the consistency and reliability of the results. To ensure dependability and replicability, scientists should actively track growth, migration, and gene/protein profiles at specific passage numbers [58]. Prolonged culture can lead to reduced or altered key functions, rendering cell lines unreliable models of their original source material [59].…”
