“…Data related to Tuberous Sclerosis and Epilepsy were obtained from the Gene Expression Omnibus (GEO) database ( 12 ). Specifically, we downloaded the datasets GSE16969 ( 13 ) and GSE62019 ( 14 ), as well as single cell sequencing data from GSE201048 ( 15 ) ( Supplementary Table 1 ). Lipid metabolism related pathways (LMRPs) were also sourced from the following PubMed articles: PMID35222371 ( 16 ), PMID36091041 ( 17 ), PMID36860853 ( 18 ), and PMID37469520 ( 19 ) ( Supplementary Table 2 ).…”

“…The above-mentioned studies, which merely demonstrate changes in the peripheral abundance of Tregs among patients with epilepsy, subsequently prompted researchers to examine the effects of this intriguing immune cell type within the CNS of epilepsy patients, where the majority of tissue damage and inflammation is localized. A bioinformatics study by Kan et al, 60 on a microarray dataset obtained using cortical samples from a small group of patients who had undergone resection of the seizure focus as treatment for focal cortical dysplasia, suggested slightly higher levels of brain infiltrated Tregs in the group with focal cortical dysplasia. Also, Xu et al noticed a substantial number of infiltrated CD4 + , CD8 + , and γδ T cells in the epileptic lesions of drug-resistant pediatric epilepsy patients suffering from focal cortical dysplasia and encephalomalacia.…”

“…67 In conclusion, several studies observed a direct increase in the number of Tregs in the brain of patients with epilepsy. 22,33,60 Some studies also demonstrated an increase in IL-10, a cytokine secreted by Tregs. 29,32,62 Moreover, the number of Tregs negatively correlated with seizure burden in human subjects.…”
