Pathophysiologic and Immunologic Changes in a Canine Endotoxemia over a Period of 24 Hours

Abstract: ABSTRACT. In this study, the pathophysiologic and immunologic parameters from a 24-hr of canine endotoxemia model by lipopolysaccharide (LPS) infusion were evaluated. For that, twelve healthy beagles received a continuous 24-hr IV infusion of low dose LPS (10 µg/kg/h, from Escherichia coli serotype O111:B4) dissolved in saline. Complete blood counts and serum biochemical analysis as well as histopathologic examination were performed to assess pathophysiologic changes such as neutrophil migration and organ inju… Show more

“…It is known that the secretion of pro-inflammatory cytokines, such as IL-1β, TNF-α, and IL-6, is increased in the initial stage of sepsis, followed by activation of anti-inflammatory mechanisms 20-22. In a recent study from an experimental sepsis model, the pattern of serum cytokine levels was associated initial leukopenia and prolonged thrombocytopenia 23. Therefore, AKI in sepsis is not solely affected by hemodynamic changes, lower WBC and platelet counts might indicate more severe immunologic responses subsequently leading to AKI.…”

“…SIRS is largely mediated by proinflammatory cytokines, and a SIRS‐like clinical picture can be induced by injection of cytokines such as tissue necrosis factor α (TNF‐α) . Infusion of lipopolysaccharide to dogs causes a rapid increase in TNF‐α concentrations that peaks within 3 hours of the start of administration . Serum concentrations of TNF‐α have been correlated with morbidity and mortality in humans with meningococcal meningitis and septicemia …”

“…Endotoxin entry into circulation from gram-negative bacteria, which are most commonly isolated in sepsis, gives rise to multiple organ failure, severe sepsis, and endotoxemia (1). Thus, experimental intravenous administrations of endotoxin (lipopolysaccharide, LPS) in humans (2), dogs (3)(4)(5), ovines (6), and calves (7) have been done to emulate sepsis (7,8).…”
