Patterns of cyclooxygenase-1 and -2 expression in human gliomas in vivo

Abstract: Cyclooxygenases (COX, prostaglandin endoperoxide synthases, PGG/H synthases) are potent mediators of inflammation. While COX-1 is constitutively expressed in a wide range of tissues, COX-2 is cytokine inducible. Although COX-1 expression is observed in normal tissue, enhanced COX-2 expression has been attributed a key role in the development of edema, impeding blood flow and immunomodulation observed in pathologically altered tissues. Here, we have analyzed the expression of COX-1 and COX-2 in 50 gliomas and 1… Show more

“…The expression of COX-2, as well as the levels of its reaction products, the prostaglandins, are frequently found elevated in gliomas. [39][40][41][42] In addition, high COX-2 expression is associated with clinically more aggressive tumors, e.g., glioblastoma multiforme, and is a strong predictor of poor survival. 43 It is therefore possible that the inhibition of COX-2 activity in such cells might be beneficial for the management of these tumors.…”

“…6 COX-2 expression has also been detected in colorectal, gastric, esophageal, and lung carcinomas, as well as in brain tumors. [7][8][9][10][11][12][13] COX-2 has been reported to be important in the relationship between increased prostaglandin synthesis and the development of glioma and its progression.…”

“…Cyclooxygenase (COX), an enzyme important for the synthesis of prostaglandins, is present in three isoforms. COX-1 shows low or constitutive expression in a variety of tissues (5,23,44) . COX-2 is highly inducible in response to cellular activation by hormones, proinflammatory cytokines, growth factors and tumor promoters (41,44) .…”
