PcG recruitment by the YY1 REPO domain can be mediated by Yaf2

Abstract: The Polycomb Group (PcG) complex of transcriptional repressors is critical for the maintenance of stage-specific developmental gene expression, stem cell maintenance and for large-scale chromosomal dynamics. Functional deficiency of a single PcG gene can severely compromise PcG function, leading to developmental defects, embryonic lethality, or a number of malignancies. Despite the critical nature of PcG proteins, the mechanisms by which these complexes mediate their effects are relatively uncharacterized. Nea… Show more

“…The mouse PRE, PRE-kr, was also found to have a YY1 DNA-binding motif within it (17). Other work has shown recruitment of PcG proteins to DNA by YY1 and found this to require the REPO domain (34). YY1 can also bind Xist RNA and serve as a docking protein for PRC2 at the inactive X chromosome (35), indicating that YY1 plays several roles in targeting PcG function.…”

“…To our knowledge, up to date, none of PRC2 core components were possessed the sequence-specific DNAbinding activity, the exact mechanism of EZH2 recruiting to target gene is still unrevealed (Muller and Kassis, 2006;Wilkinson et al, 2010). Recently, Herranz et al (2008) found that zinc-finger factor Snail could recruit PRC2 complex to the E-cadherin promoter (Herranz et al, 2008).…”

“…The potential to associate with a DNA binding protein underlies a proposed role as recruiter of PcG complexes to their targets. However, despite some evidence for such an activity (62,63), chromatin association studies in ES cells failed to show YY1 colocalization with PcG targets (33). Importantly, in ES cells RYBP is also part of protein complexes containing core transcription factors of the pluripotent network (Pou5f1/Oct4) (57,60), and ES cell lines cannot be established from RYBP-deficient early embryos (41).…”
