PD-L1/PD-1 and CTLA-4 Expression in Equine Penile Squamous Cell Carcinomas

Abstract: In horses, penile squamous cell carcinomas (epSCCs) are among the most common cutaneous neoplastic lesions. These tumors usually arise in benign lesions such as viral plaques and papillomas frequently induced by Equus caballus papillomavirus type 2 (EcPV2) infection. In the last decade, the introduction of immune checkpoint inhibitors (ICI) for the treatment of human cancers has demonstrated promising results. Among the most commonly targeted pathways, there is PD-1/PD-L1 and CTLA-4. The aim of this study is t… Show more

“…EcPV2 detection was assessed in 100 ng of DNA sample. In particular, L1, E2, E6, and E7 amplification by the real-time PCR assay was performed applying previously described protocols [10,18,19] and using the amplification of the equine beta-2-microglobulin (B2M) gene [10,20] as the control for DNA integrity (primer and probes sequences are reported in Table 1). Moreover, internal controls (block blanks, extraction blanks, and positive controls) were used for each analytical session.…”

“…Anti-bovine PD-L1 antibody shows cross-reactivity with porcine PD-L1 [ 67 ] and canine PD-L1 [ 68 ]. In addition, cross-reactive anti-human PD-1, PD-L1, or PD-L2 antibodies were used in canine [ 69 ], equine [ 70 ], and feline tissues [ 71 ].…”

“…The examination of 20 equine penile squamous cell carcinomas revealed PD-L1 mRNA and protein in three cases and one case, respectively, whereas PD-1 expression was not detected [ 70 ].…”

“…Considering the existent evidence regarding the positive clinical effects of PD-1/PD-L1 blockade in the treatment of melanoma, it is pertinent to assess whether equine melanomas are potential candidates for this therapy. Although some articles have been published, according to the authors’ knowledge, the literature regarding PD-L1 expression in equine tumors is scarce and the sample dimension in the published articles is quite small, compromising the interpretation of the results [ 53 , 54 , 55 ].…”
