The study aims to discuss the challenges associated with treating prostate cancer (PCa), which is known for its complexity and drug resistance. It attempts to find differentially expressed genes (DEGs), such as those linked to anoikis resistance and circulating tumor cells, in PCa samples. This study involves analyzing the functional roles of these DEGs using gene enrichment analysis, and then screening of 102 bioactive compounds to identify a combination that can control the expression of the identified DEGs. In this study, 53 DEGs were identified from PCa samples including anoikis-resistant PCa cells and circulating tumor cells in PCa. Gene enrichment analysis with regards to functional enrichment of DEGs was performed. An inclusive screening process was carried out among 102 bioactive compounds to identify a combination capable of affecting and regulating the expression of selected DEGs. Eventually, gastrodin, nitidine chloride, chenodeoxycholic acid, and bilobalide were selected, as their combination demonstrated ability to modulate expression of 50 out of the 53 genes targeted. The subsequent analysis focused on investigating the biological pathways and processes influenced by this combination. The findings revealed a multifaceted and multidimensional approach to tumor regression. The combination of bioactive compounds exhibited effects on various genes including those related to production of inflammatory cytokines, cell proliferation, autophagy, apoptosis, angiogenesis, and metastasis. The current study has made a valuable contribution to the development of a combination of bioactive natural compounds that can significantly impede the development of treatment resistance in prostate tumor while countering the tumors’ evasion of the immune system. The implications of this study are highly significant as it suggests the creation of an enhanced immunotherapeutic, natural therapeutic concoction with combinatorial potential.

Background: Pediatric leukemia poses considerable burdens on caregivers, precipitating psychological distress and financial strain. Caregivers may experience cognitive, emotional, and behavioral shifts. The adverse event, such as the COVID-19 pandemic, may exacerbate their psychological distress. The lived experience of parental psychological resilience in the context of pediatric leukemia caregiving warrants greater scrutiny during the COVID-19 pandemic period. This investigation delves into the lived experience of the psychological resilience dynamics among parents of children with leukemia. Methods: Employing phenomenological methodology, semi-structured interviews were undertaken with parents of pediatric leukemia patients at varying stages of the disease, admitted to a tertiary care pediatric hematology unit in Sichuan Province between March and September 2021. Data analysis was performed using NVivo 11.0 software alongside Colaizzi's phenomenological analysis technique. Results: The study encompassed twenty-four primary caregivers of children with leukemia, including 19 mothers and 5 fathers, with an age span of 28 to 48 years, a mean (SD) of 34.67 (5.39). Analysis revealed five primary themes and seven sub-themes, encompassing: complex emotional trajectories marked by simultaneous negative and positive feelings; the considerable burden of care juxtaposed with inadequate coping mechanisms; a pronounced eagerness for caregiving knowledge contrasted with a deficit in essential training; curtailed social engagement and challenges in resuming normalcy; and a pronounced need for psychological support. Conclusions: The emotional landscape for parents caring for children with leukemia is characterized by a spectrum of lived experiences and emotions, both positive and negative. These emotional shifts are shaped by diverse factors. Positive emotions have the potential to bolster resilience in parents, enabling them to navigate adversity, surmount challenges, and proactively manage the psychological strains and stresses engendered by such crises. Mitigating caregiving demands and augmenting social and psychological support are crucial for enhancing life quality and wellbeing among these parents.

Genomic instability is one of the main drivers of tumorigenesis and the development of hematological malignancies. Cancer cells can remedy chemotherapeutic-induced DNA damage by upregulating DNA-repair genes and ultimately inducing therapy resistance. Nevertheless, the association between the DNA-repair genes, drug resistance, and disease relapse has not been well characterized in acute lymphoblastic leukemia (ALL). This study aimed to explore the role of the DNA-repair machinery and the molecular mechanisms by which it is regulated in early- and late-relapsing pediatric ALL patients. We performed secondary data analysis on the Therapeutically Applicable Research to Generate Effective Treatments (TARGET)—ALL expansion phase II trial of 198 relapsed pediatric precursor B-cell ALL. Comprehensive genetic and epigenetic investigations of 147 DNA-repair genes were conducted in the study. Gene expression was assessed using Microarray and RNA-sequencing platforms. Genomic alternations, methylation status, and miRNA transcriptome were investigated for the candidate DNA-repair genes. We identified three DNA-repair genes, ALKBH3, NHEJ1, and PARP1, that were upregulated in early relapsers compared to late relapsers (p < 0.05). Such upregulation at diagnosis was significantly associated with disease-free survival and overall survival in precursor-B-ALL (p < 0.05). Moreover, PARP1 upregulation accompanied a significant downregulation of its targeting miRNA, miR-1301-3p (p = 0.0152), which was strongly linked with poorer disease-free and overall survivals. Upregulation of DNA-repair genes, PARP1 in particular, increases the likelihood of early relapse of precursor-B-ALL in children. The observation that PARP1 was upregulated in early relapsers relative to late relapsers might serve as a valid rationale for proposing alternative treatment approaches, such as using PARP inhibitors with chemotherapy.
