Peg-Bovine Hemoglobin: Safety en a Canine Dehydrated Hypovolemic-Hemorrhagic Shock Model

Abstract: An initial evaluation of PEG-bHb was performed using a modified hypovolemic shock model. PEG-bHb had a substantially longer intravascular half-life than native Hb and no measurable hemoglobinuria was observed in the canine. PEG-bHb allowed successful resuscitation with an oxygen carrying capacity of 14-22% over that of lactated Ringer's solution.

“…Surface lysine residues on purifi ed bovine Hb were conjugated with 5000 molecular weight PEG (Nho et al, 1992). Th e PEG-Hb solution was bubbled with CO to convert greater than 80% of the PEG-Hb to PEG-COHb before storage.…”

“…Sanguinate is a purified bovine Hb manufactured by Prolong Pharmaceuticals (Piscataway, NJ, USA) and is conjugated with 5000 molecular weight of PEG residues on the surface lysines and is ligated to CO [57]. The unligated form of this HBOC has a high oxygen affinity (a P 50 of approximately 11 mmHg).…”

“…A Phase I/II study using polymerized bovine hemoglobin (HBOC-201, Hemopure) was carried out in adult patients with sickle cell disease who were not at a crisis at the time of the study [57]. In this investigation, 18 adults with SCD who were asymptomatic at the time were enrolled for this study.…”
