Penetration studies of topically applied substances: optical determination of the amount of stratum corneum removed by tape stripping

Abstract: Tape stripping is a standard measuring method for the investigation of the dermatopharmacokinetics of topically applied substances using adhesive films. These tape strips are successively applied and removed from the skin after application and penetration of topically applied substances. Thus, layers of corneocytes and some amount of topical applied substances are removed. The amount of substances and the amount of stratum corneum removed with a single tape strip has to be determined for the calculation of the… Show more

“…Since a large discrepancy of skin penetration existed among the various buffers, the partitioning or permeation process via skin but not the release process was the rate-limiting step for Se-L-M. Tape stripping is a simple standard technique for evaluating the function of the SC in percutaneous absorption studies [33] . Se-L-M in pH 4 buffer showed the highest enhancement of skin permeation across SC-stripped skin compared to the other buffer systems, suggesting that diffusion through the SC layer was important for permeation in a cationic form.…”

“…Tape stripping was performed 10 min after application and pH SC was measured after every 10 tape strippings. In total, 100 tape strippings were performed to remove the complete SC [16,17]. Changes in pH SS over time were measured without removal of the SC on the other arm.…”

“…Tape stripping removed SC partially or totally, and it is a simple standard technique for the establishment of skin barrier-damaged model. 30 However, NP penetration through tape stripped skin did not always lead to marked penetration enhancement. It varied qualitatively in magnitude from none [31][32][33][34] to some NPs detected.…”

“…29,30 To investigate the permeability of NPs in barrier-damaged skin, we tape stripped samples mimicking barrier disruption. Tape stripping removed SC partially or totally, and it is a simple standard technique for the establishment of skin barrier-damaged model.…”
