Penicillin-Binding Protein 1A, 2B, and 2X Alterations in Canadian Isolates of Penicillin-Resistant Streptococcus pneumoniae

Abstract: Alterations within the penicillin-binding domain of penicillin-binding protein (PBP) genes pbp1a, pbp2b, and pbp2x were determined for 15 Canadian isolates of Streptococcus pneumoniae. All penicillin-nonsusceptible S. pneumoniae isolates showed a variety of PBP 2X substitutions and contained a Thr445-Ala change after the PBP 2B SSN motif. Only isolates for which penicillin MICs were >0.5 g/ml had PBP 1A alterations near the STMK and SRN motifs. Sequence analysis revealed identical PBP 1A, PBP 2B, and PBP 2X su… Show more

“…These findings suggest that antibiotic pressure is capable of promoting genetic rearrangement in S. pneumoniae. The alteration ratios for PBPs have been reported by a few investigators (9,21), but these pieces of information were restricted to the TP domains of PBPs. Clinical practitioners in Japan prefer to use cephalosporins instead of penicillins for the treatment of community-acquired infections, and the prevalence of cephalosporin-resistant S. pneumoniae strains in Japan has been increasing year by year (26).…”

“…However, some pneumococcal strains did not belong to these classes and cannot be detected by class-specific PCR assays (24,25). Genetic analysis of pbp2b sequences in penicillin-resistant strains were reported from Japan, Spain, Iceland, Canada, France, the United Kingdom, Papua New Guinea, South Africa, and the United States (7,10,11,17,19,20,21,25). In Korea, the pbp2b gene sequences of multidrug-resistant S. pneumoniae isolates were reported (23).…”

“…Approximately 1-kb fragments of pbp1a, pbp2x, and pbp2b encompassing the penicillin-binding domain of each gene were PCR amplified as described by Nichol et al (17) and sequenced by ACGT (Wheeling, IL). Competition assays were performed with a protocol based on the work of Sifaoui et al (20), using Bocillin-FL (Invitrogen, Carlsbad, CA).…”
