Pentraxin family of proteins interact specifically with phosphorylcholine and/or phosphorylethanolamine

Abstract: Pentraxins are a family of serum proteins characterized by five identical subunits that are noncovalently linked. The two major types of pentraxins are C-reactive protein (CRP) and serum amyloid P component (SAP). CRP proteins are identified by their calcium-dependent interaction with phosphorylcholine. This study showed that SAP also bound to phosphorylated compounds but had a high specificity for phosphorylethanolamine. Thus, human CRP and SAP show high specificity that is complementary for the related compo… Show more

“…Similar to our findings for CRP, the C4BP binding site for SAP is located in the central core region of the protein, suggesting an analogous interaction between the two pentraxins and C4BP. Contrary to what we have found in the present study, previously published data indicated that SAP and CRP from rat interact with C4BP while human CRP is unable to bind human C4BP (29). The initial study did not investigate complexes between C4BP and CRP in a situation when one of the ligands is immobilized.…”

“…In the presence of calcium ions, SAP can bind to several ligands such as amyloid fibrils of any type (4), agarose (8), heparin and dermatan sulfate (9), C1q (10,11), C4 binding protein (12,13), laminin (14), type V collagen (15), and phosphoethanolaminecontaining compounds such as phosphatidylethanolamine (16,17), DNA (18), chromatin (19,20), and histones (21,22).…”

“…Because E-LDL was not visible in the photograph of the stained gel, we performed Western blotting using anti-ApoB Abs. Western blotting showed the presence of E-LDL in these fractions (lanes [11][12][13][14]. Thus, in the presence of PEt, binding of CRP to fluidphase E-LDL was enhanced.…”
