Peptide Selection of MMP-1 for Electrochemical Sensing with Epitope-Imprinted Poly(TPARA-co-EDOT)s

Abstract: Instead of molecularly imprinting a whole protein molecule, imprinting protein epitopes is gaining popularity due to cost and solubility issues. Belonging to the matrix metalloproteinase protein family, MMP-1 is an interstitial collagenase that degrades collagen and may be involved in cell migration, cell proliferation, the pro-inflammatory effect, and cancer progression. Hence, it can serve as a disease protein biomarker and thus be useful in early diagnosis. Herein, epitopes of MMP-1 were identified by scree… Show more

“…Metalloproteinase MMP-1 can be regarded as a disease biomarker, which is meaningful in early diagnosis, so it is of great significance for the selective separation of MMP-1. Epitope peptide was prepared by cleaving MMP-1 in silico with trypsin, and peptide fragments were obtained [137]. The EIPs were synthesized with Metalloproteinase MMP-1 can be regarded as a disease biomarker, which is meaningful in early diagnosis, so it is of great significance for the selective separation of MMP-1.…”

“…Figure 1(c) plots the CVs for the doping of various concentrations of MoS 2 nanosheets with the same mole ratio of TPARA to EDOT. 20 The current densities in the first cycles of both MoS 2 -doped (peptide A imprinted polymers, pAIPs) and undoped pAIPs revealed equal conductivity. However, the additional MoS 2 nanosheets inhibit the increase of current densities in the MoS 2 -doped pAIPscoated layer during electropolymerization.…”

“…Clearly, all of the first cycles are very similar; however, the increase in the current at the peak oxidation voltage is lower for a higher MoS 2 doping concentration, as shown in Figures S1(a)–(d). Figure (c) plots the CVs for the doping of various concentrations of MoS 2 nanosheets with the same mole ratio of TPARA to EDOT . The current densities in the first cycles of both MoS 2 -doped (peptide A imprinted polymers, pAIPs) and undoped pAIPs revealed equal conductivity.…”
