Percutaneous US-Guided Implantation of Vx-2 Carcinoma into Rabbit Liver: A Comparison With Open Surgical Method

Abstract: Purpose-To evaluate technical feasibility and experimental usefulness of percutaneous USguided implantation of Vx-2 carcinoma in rabbit liver.Materials and Methods-Forty-eight New Zealand White male rabbits were used. Solid tumor mass of Vx-2 carcinoma was harvested from carrier rabbit, and minced tumor cells were implanted. Twenty-four rabbits underwent percutaneous US-guided tumor implantation, and the same number of rabbits underwent open laparotomy tumor implantation. Tested parameters included technical s… Show more

“…However, tumor seeding occurred more often (20.8%) in the ultrasound-guided approach than in their laparotomy controls (8.3%). In addition, there was thoracoabdominal cancer cell seeding in 20.8% of cases, and also in the peritoneal cavity in 8.3% of the cases 29 . A percutaneous technique has been previously described for the inoculation of human hepatocytes in immunodeficient mice 30 .…”

“…Lee et al 29 have described a percutaneous ultrasound-guided approach to inoculate Vx-2 carcinoma cells in the liver of rabbits. This technique resulted in a high success rate (100%) and was less invasive than an open laparotomy method.…”

“…Each pre-anaesthetised rabbit was placed in a supine position, shaved in the subxyphoid area, and prepped in a sterile fashion following methods previously described [43,44]. Using real-time ultrasound (US) guidance (7.5-MHz linear transducer, Aloka, Wallingford, CT), percutaneous implantation of two aliquots (0.1 mlL) of VX2 tumour pieces (previously excised from the thigh of a rabbit VX2 tumour carrier) was performed via a 21-gauge angiographic catheter.…”

“…Anesthesia was induced with 5% isoflurane (Hospira, Warwickshire, England) and 95% oxygen (Air Gas, Salem, NH) and then sustained with 2.5% isoflurane and 97.5% oxygen at a rate of 3.5 L/min, with intravenous bolus injections of 0.25 mL propofol (10 mg/mL) (APP Pharmaceuticals, Schaumburg, Ill) via a marginal ear vein. Chunks of VX2 tumor were excised from the hind legs of the carriers, minced, and surgically implanted into the left lobe of the liver as previously described (28). The liver tumors were monitored with ultrasonography (SonoSite 180 PLUS; SonoSite, Bothell, Wash) and allowed to grow for 12-15 days after tumor implantation to reach dimensions of 2-3 cm.…”
