Pericyte Loss in Diseases

Abstract: Pericytes are specialized cells located in close proximity to endothelial cells within the microvasculature. They play a crucial role in regulating blood flow, stabilizing vessel walls, and maintaining the integrity of the blood–brain barrier. The loss of pericytes has been associated with the development and progression of various diseases, such as diabetes, Alzheimer’s disease, sepsis, stroke, and traumatic brain injury. This review examines the detection of pericyte loss in different diseases, explores the … Show more

“…The initial occurrence associated with AD is the accumulation of Aβ, which can lead to cerebral amyloid angiopathy . The buildup of Aβ surrounding blood vessels has the potential to cause pericyte death and altered astrocyte morphology, consequently compromising BBB integrity (Figure B) . Another major pathology hallmark in AD is the presence of tau deposits .…”

“…In the brain and retina, pericyte-to-endothelial cell ratio is 1:1, and pericytes are important components of the blood-brain barrier and blood-retina barrier. 66 In the lungs, pericyte-to-endothelial cell is 1:7-1:9 and pericytes are involved in optimal gas exchanges. 66 In the kidney, the ratio is 1:2.5, and changes in renal vascular permeability amplify renal structural and functional disorders.…”

“…PCs cover the outer side of capillaries and can regulate blood flow by controlling the diameter of the capillaries. In addition, PCs can also participate in clearing toxic proteins and forming tight junctions [ 40 ]. Astrocytes are the most widely distributed cells in the central nervous system.…”
