Perifollicular Transgenic Expression of Human Interleukin-1 Receptor Antagonist Protein following Topical Application of Novel Liposome-Plasmid DNA Formulations In Vivo

Abstract: Expression plasmid DNA for the human interleukin-1 receptor antagonist (IL-1ra) protein was formulated with nonionic:cationic (NC) liposomes or phosphatidylcholine:cationic (PC) liposomes and applied to the auricular skin of hamsters in single- and multiple-dose protocols. Confocal microscopy identified delivery of plasmid DNA proximal to perifollicular cells, and successful transfection of perifollicular cells was identified by immunohistochemistry and ELISA. Skin treated for 3 days with the NC liposomes had … Show more

“…There are some reports describing that liposomes have been used to enhance transfollicular and transdermal delivery of the low-/high-molecular weight and hydrophilic/lipophilic compounds (Fleisher et al, 1995;Li and Hoffman, 1996;Niemiec et al, 1997;Niemiec et al, 1995;Seiberg et al, 1997). However, we have confirmed that in vivo permeation of liposomes from the hair follicles into dermis/epidermis was not appeared under passive condition ( Supplementary Fig.…”

“…1 To accomplish optimal and prolonged gene expression the delivery vehicle plays an important role in protecting DNA from physical, chemical, and enzymatic destruction. Delivery vehicles that bind to DNA and facilitate transport to cellular targets include peptides, 2 glycopeptides, 3 glycosylated polymers, 4,5 cationic lipids, [6][7][8][9][10] liposomes, 11,12 and PLGA microspheres. 13 During fabrication of a delivery system the structure of DNA can be altered by shear stress to convert supercoiled plasmid to open circular, linear, or even fragmented DNA.…”

“…Expression of transgenic proteins could correct pertubances in cytokine production, the cause of several diseases affecting the skin. Liposomal delivery of IL-1receptor antagonist and expression of protein in the cells could inhibit the pathological effects of the endogenous IL-1 in skin cells, provided that the levels of the antagonist are 10-100 fold higher than the cytokine levels [101].…”
