Periodontal Ligament Cell Sheets and RGD-Modified Chitosan Improved Regeneration in the Horizontal Periodontal Defect Model

Abstract: Objective The aim of this study was to examine the potential of periodontal ligament (PDL) cells sheet and arginine-glycyl-aspartic acid (RGD)-modified chitosan scaffold for periodontal tissue regeneration in horizontal periodontal defect model. Materials and Methods PDL cell cytotoxicity was tested with 3–[4,5- dimethylthiazol-2yl]–2,5-diphenyl-2H-tetrazolium bromide assay. Cell migration toward the chitosan-based materials was analyzed with trans-well migration assay. Horizontal periodontal defect model was … Show more

“…These properties and their high availability also reveal the potential usefulness of PDLCs as cell therapy in periodontal disease, regenerative medicine, autoimmune diseases, and inflammatory diseases ( 58 , 59 ). Autologous periodontal membrane cell transplantation can be used to regenerate periodontal tissue defects ( 60 62 ). Our study suggests that PDLCs treated with IL-7 may have more immunosuppressive properties and enhanced ability of periodontal tissue regeneration and repair, which may contribute to the maintenance of periodontal tissue health in future clinical practice and the prevention and treatment of periodontal tissue defects caused by inflammation.…”

“…39 Further, periodontal ligament cells sheet and arginine-glycyl-aspartic acid-modified chitosan scaffold for periodontal tissue regeneration showed biocompatibility and high cell proliferation in a horizontal periodontal defect model. 40 On the other hand, chitosan hydrogels (chitosan-H-propolis, chitosan-H-propolis-nystatin, and chitosan-H-nystatin) treated dentine gave significantly high shear bond values by their swelling properties and antioxidant effect. 41 Besides, the anticarious mats of chitosan fibers containing Garcinia mangostana extract have shown promising results to prevent dental caries.…”

“…However, they are prone to other infectious diseases, less control over post-operative trauma, and high cost of maintenance [ 138 ]. Amir et al demonstrated potential use of PDL cell sheets and RGD chitosan scaffold for periodontal regeneration including PDL, AB, and CM in M. nemestrina monkey model [ 142 ].…”
