PET evaluation of light-induced modulation of microglial activation and GLP-1R expression in depressive rats

Abstract: Light therapy has been accepted as a promising therapeutic choice for depression. Positron emission tomography (PET) combined with specific radiotracers has great benefits for revealing pathogenesis and developing therapeutics. This study aimed to investigate the influences of light therapy on microglial activation and glucagon-like peptide-1 receptor (GLP-1R) expression in the brain of depressive rats using [18F]DPA-714 and [18F]exendin-4 PET. The results showed that chronic unpredictable mild stress (CUMS)-i… Show more

“…These regions may be linked to the onset of depressive symptoms in humans [28]. The observed temporospatial neuroinflammation-related TSPO-PET imaging data generated herein coincide well with data from prior IHC-based analyses of inflammation-induced and stress-related depression models [29]. Moreover, these data align with recent evidence regarding increased TSPO levels in the brains of MDD patients relative to healthy age-matched controls as detected via [18F] FEPPA PET imaging [30].…”

“…The data from the present study added new proofs that oxidative stress induced by immunoinflammation played an important role in depressive behaviors in hypothyroid rats. This is not a unique instance—in another study, DPA-714 uptake increased in 11 regions of interest in the brain of depressed rats [ 17 ]. This suggests that depression-like behaviors associated with hypothyroidism in rats may have a common mechanism with depression.…”

“…However, data on the changes in GLP-1R levels in other CNS disorders accompanied by microglial activation are contradictory. For example, the expression of GLP-1R in Alzheimer's disease, experimental autoimmune encephalomyelitis and depression [46][47][48] was found to be decreased in the brain. The differences in GLP-1R expression in activated microglia might be due to the different regions of the CNS and the various methods for activating microglia.…”
