PET Study of Sphingosine-1-phosphate Receptor 1 Expression in Response to S. aureus Infection

Abstract: Sphingosine-1-phosphate receptor 1 (S1PR1) plays a crucial role in infectious diseases. Targeting S1PR1 provides protection against pathogens, such as influenza viruses. This study is aimed at investigating S1PR1 in response to bacterial infection by assessing S1PR1 expression in S. aureus-infected mice. A rodent local muscle bacterial infection model was developed by injecting S. aureus to the lower hind limb of Balb/c mice. The changes of S1PR1 expression in response to bacterial infection and blocking treat… Show more

“…1 and have identical binding properties and brain washout pharmacokinetics in nonhuman primates. 23 In addition, we also reported other potent 18 F-labeled S1PR1 radiotracers, including [ 18 F]TZ4877 [29][30][31][32][33] and [ 18 F]TZ82112, 34,35 contain a terminal alcohol as the hydrophilic head group (Fig. 1).…”

“…Positron emission tomography (PET) imaging of S1PR1 in humans provides a new methodology for the early detection and monitoring therapeutic efficacy. We reported several S1PR1-selective PET radioligands, , including [ 11 C] CS1P1 , [ 18 F] FS1P1 , [ 18 F] TZ4877 , and [ 18 F] TZ82112 , for imaging changes in S1PR1 expression in the brain (Figure ). [ 11 C] CS1P1 and [ 18 F] FS1P1 share the same molecular structure but are labeled at different positions with different isotopes.…”

“…1) has an IC 50 value of 14.01±0.05 nM for S1PR 1 and selectivity over other S1PR subtypes (> 100-fold over S1PR 2 − 5 18 ), and in rat is well-described by the 2-tissue compartment model (2TCM) with reproducible estimates of volume of distribution (V T ), with no evidence of radiolabeled metabolites penetrating the brain 17 . [ 18 F]TZ4877 also exhibited elevated uptake in a mouse model of bacterial infection 19 and rat model of experimental autoimmune encephalomyelitis (EAE) 17 . These data indicate that [ 18 F]TZ4877 has potential for imaging S1PR 1 in human subjects.…”
