Pex15p of Saccharomyces cerevisiae Provides a Molecular Basis for Recruitment of the AAA Peroxin Pex6p to Peroxisomal Membranes

Abstract: The gene products (peroxins) of at least 29 PEX genes are known to be necessary for peroxisome biogenesis but for most of them their precise function remains to be established. Here we show that Pex15p, an integral peroxisomal membrane protein, in vivo and in vitro binds the AAA peroxin Pex6p. This interaction functionally interconnects these two hitherto unrelated peroxins. Pex15p provides the mechanistic basis for the reversible targeting of Pex6p to peroxisomal membranes. We could demonstrate that the N-ter… Show more

“…5). Among these five mutants, two groups can be distinguished based on the pattern of ubiquitinated Pex5p, and the peroxins deficient within a group are functionally related (18,22). The first group consists of the pex4⌬ and pex22⌬ mutants, whereas the second group comprises pex1⌬, pex6⌬, and pex15⌬.…”

“…Two peroxins, Pex1p and Pex6p, are known as ATPase associated with various cellular activities (AAA) ATPases (16,17). In Saccharomyces cerevisiae they partially associate with the membrane via the integral membrane protein Pex15p (18,19). AAA ATPases have been suggested to function in the dissociation of protein complexes and protein unfolding (for review, see Refs.…”

“…Immunoblot analysis was performed according to Harlow and Lane (34) with polyclonal rabbit antibodies raised against Pex7p (35), Pex13p (36), Pex14p (9), Fox3p (37), Pcs60p (38), Gpd1p, Porin, and Kar2p (39), Fbp1p (40), protein A (Sigma, Munich, Germany), and GFP-GST (41). Primary antibodies were detected with an IRDye 800CW goat anti-rabbit IgG secondary antibody (LI-COR Bioscience, Bad Homburg, Germany).…”
