Pharmacogenetics of platinum-based chemotherapy: impact of DNA repair and folate metabolism gene polymorphisms on prognosis of non-small cell lung cancer patients

Abstract: Chemotherapy based on platinum compounds is the standard treatment for NSCLC patients with EGFR wild type, and is also used as second line in mutated EGFR patients. Nevertheless, this therapy presents poor clinical outcomes. ERCC1, ERCC2, XRCC1, MDM2, MTHFR, MTR, and SLC19A1 gene polymorphisms may contribute to individual variation in response and survival to platinum-based chemotherapy. The aim of this study was to investigate the influence of these polymorphisms on response and survival of NSCLC patients tre… Show more

“…In the current study, the MDM2 rs1690924 mutation was significantly associated with grade 3-4 GI toxicity, and the carriers of the TT + TC genotype had a markedly increased risk of grade 3-4 GI toxicity than the carriers of the CC genotype did. To date, only one study found an association between this polymorphism and overall survival (41). However, the function of MDM2 rs1690924 polymorphism is unknown.…”

“…There are two polymorphisms of XRCC1 , rs25487 (Arg399Gln) and rs1799782 (Arg194Trp), that are believed to change the DNA repair activity and affect the sensitivity of tumor cells to platinum drugs 14 . Thus, they have been widely investigated for the association with response to platinum-based chemotherapy in NSCLC patients 15 17 , mostly in Asian populations 18 22 . However, the results remain inconclusive.…”

“…Previous study also revealed that rs25487 GG genotype could decrease the function of XRCC1, and those with rs25487 GG genotype showed significantly better ORR; thus, this genotype could act as predictive factors in NSCLC patients treated with platinum-based chemotherapy. 39 Therefore, in our paper, we observed that patients with rs3213245 minor C allele tended to have good response to chemotherapy, although there are no significant differences.…”
