Pharmacogenomic Characterization and Isobologram Analysis of the Combination of Ascorbic Acid and Curcumin—Two Main Metabolites of Curcuma longa—in Cancer Cells

Abstract: Curcuma longa has long been used in China and India as anti-inflammatory agent to treat a wide variety of conditions and also as a spice for varied curry preparations. The chemoprofile of the Curcuma species exhibits the presence of varied phytochemicals with curcumin being present in all three species but AA only being shown in C. longa. This study explored the effect of a curcumin/AA combination on human cancer cell lines. The curcumin/AA combination was assessed by isobologram analysis using the Loewe addit… Show more

“…Variants in FAM86HP (Family With Sequence Similarity 86 Member H, Pseudogene) have been reported to be associated with BMI-adjusted waist-hip ratio [49]. In the pathway analysis (Fig 5), we observed that FAM86HP has an indirect interaction with TGM2 (Transglutaminase 2), a stress-response gene [50] involved in mRNA metabolism [51] which has been reported to be upregulated during in ammation [50]. It is interesting to note that TGM2 was over expressed in individuals receiving chemo & radiation therapy, suggesting its involvement in sensitivity to radiation [52,53].…”

“…Isobolograms were generated and the results of isobolographic analysis for various GT combinations are presented in Figure a–d. It is a graphical representation composed of individual drug doses and a straight line of additivity is employed to determine the drug interactions, if the experimental value below this line indicates synergism, above it shows antagonistic interaction, and on the line of additivity demonstrates additive effect (Ooko, Kadioglu, Greten, & Efferth, ).…”
