Pharmacokinetic Alteration in Rats of Recombinant Interleukin-2 (rIL-2) by Immunocomplexing with a Monoclonal Antibody against rIL-2.

Abstract: We have investigated the pharmacokinetic alteration in rats of recombinant interleukin-2 (rIL-2) by immunocomplexing with a monoclonal antibody against rIL-2. Serum rIL-2 levels after the intravenous administration of the immune complex at a dose of 100 micrograms/rat as rIL-2 were significantly higher than those after intravenous administration of rIL-2 alone at the same dose. Pharmacokinetic analysis indicated that the distribution volume of rIL-2 decreased from 74.0 to 10.3 ml/rat, while the elimination rat… Show more

“…Alternatively, cytokines in complex with anti-cytokine Abs can prolong the half-life and activity of cytokines (61–66). Supporting this, hypersensitivity can be suppressed by IL-2 in complex with IgG2b with increased stability and half-life (67).…”

“…Increases in the stability of complexed IL-2, IL-3, IL-4, IFN-␣, and IL-6 (13)(14)(15)(16)(17)(18)(19)(20)(21) and in the biological activity of the cytokine complex for IL-2, IL-3, IL-4, IL-6, IL-7, IL-8, IFN-␥, and TNF (2, 5, 16 -29) have been demonstrated. In addition to these complexes formed with Ab, recent studies established the increase of biological activity of IL-15 complexed with the IL-15 receptor chain ␣ (30,31).…”
