Pharmacokinetic Behavior and Pharmacokinetic/Pharmacodynamic Integration of Danofloxacin Following Single or Co-Administration with Meloxicam in Healthy Lambs and Lambs with Respiratory Infections

Abstract: The aim of this study was to determine the pharmacokinetics and pharmacodynamics of danofloxacin (DAN; 6 mg/kg) following subcutaneous administration alone or co-administration with meloxicam (MLX; 1 mg/kg) in healthy lambs and lambs with respiratory infections. The study was carried out using a total of four groups: HD (healthy; n = 6) and ID (infected; n = 7) groups who were administered DAN only, and HDM (healthy; n = 6) and IDM (infected; n = 7) groups who were administered DAN and MLX simultaneously. The … Show more

“…The tolfenamic acid after IV administration in rainbow trout exhibited short t 1/2ʎz with 3.47 h, which was similar to that previously reported in mammals (1.57-6.09 h, Jaussaud et al, 1992;Tekeli et al, 2020) and birds (1.51-1.95 h, Cetin et al, 2022;Turk et al, 2021b), it was shorter than reported in turtles (17.55-38.92 h, Corum et al, 2019;Raweewan et al, 2020b). Fish are heterotherms and their metabolic rates are approximately 10 times lower than mammals (Van de Pol et al, 2017). In addition, the metabolic rate of the fish is affected by the water temperature, and the metabolic rate decreases at low water temperature (Corum et al, 2023).…”

“…In plasma samples obtained from healthy fish, the plasma protein binding ratio of tolfenamic acid was evaluated using ultrafiltration method (Ural & Uney, 2021 ). Plasma collected from fish that were not treated with drug was pooled.…”

“…They work by inhibiting bacterial DNA gyrase and topoisomerase IV, preventing the normal replication of bacterial DNA and leading to irreversible damage, thus exhibiting antibacterial effects [ 4 ]. These compounds are widely used in the treatment of various bacterial infectious diseases in livestock and poultry due to their advantages such as long half-life, low plasma protein binding rate, strong bactericidal activity, ease of clinical use, and minimal side effects [ 5 ].…”

“…Moreover, the antibacterial efficacy of the three nanosystems before and after loading with danofloxacin will be evaluated. Danofloxacin is an antibiotic of the family of fluoroquinolones actives against most of the Gram-negative bacteria such as Escherichia coli, Klebsiella, and Salmonella, as well as some Gram-positive ones, including Staphylococcus aureus and Streptococcus pneumoniae [46]. Danofloxacin was selected due to its small size, broad spectrum effect and low minimal inhibitory concentration (MIC) [47].…”
