Pharmacokinetics and pharmacodynamics of mood-altering drugs in patients with cancer

Abstract: Undertreatment of depression in patients with cancer is a clinical problem requiring prompt action. Although responsive to cognitive and behavioral interventions, unipolar depression in patients with cancer often requires pharmacologic management. Because of pathologies associated with cancer and its treatment, pharmacologic outcomes are often unpredictable, necessitating careful assessment of risk factors for over- and undermedication. This article reviews important principles of pharmacokinetics and pharmaco… Show more

“…Antidepressants are very effective in the treatment of depressive symptoms and are reported to be effective in 70± 80% of the depressed population (Bell 1991;Cathcart et al 1993;Barraclough 1994;Lovejoy & Matteis 1996). The most commonly prescribed drug categories include trycyclic and more recently serotonin reuptake inhibitors (SSRls), the most commonly know ones being Prozac and Zoloft.…”

“…Neuroendocrine-immune modulation (NIM) negative feedback network is one of the important pathophysiological basis of clinical depression in patients with cancer. Depression in pancreatic cancer patients has been linked to the secretion of amines resistant to emotional excitement (7). …”
