Ceftiofur, a third‐generation cephalosporin antibiotic, is being extensively used by pet doctors in China. In the current study, the detection method was developed for ceftiofur and its metabolites, desfuroylceftiofur (DCE) and desfuroylceftiofur conjugates (DCEC), in feline plasma. Then, the pharmacokinetics studies were performed following one single intravenous and subcutaneous injection of ceftiofur sodium in cats both at 5 mg/kg body weight (BW) (calculated as pure ceftiofur). Ceftiofur, DCE, and DCEC were extracted from plasma samples, then derivatized and further quantified by high‐performance liquid chromatography. The concentrations versus time data were subjected to noncompartmental analysis to obtain the pharmacokinetics parameters. The terminal half‐life (t1/2λz) was calculated as 11.29 ± 1.09 and 10.69 ± 1.31 hr following intravenous and subcutaneous injections, respectively. After intravenous treatment, the total body clearance (Cl) and volume of distribution at steady‐state (VSS) were determined as 14.14 ± 1.09 ml hr‐1 kg‐1 and 241.71 ± 22.40 ml/kg, respectively. After subcutaneous injection, the peak concentration (Cmax; 14.99 ± 2.29 μg/ml) was observed at 4.17 ± 0.41 hr, and the absorption half‐life (t1/2ka) and absolute bioavailability (F) were calculated as 2.83 ± 0.46 hr and 82.95%±9.59%, respectively. The pharmacokinetic profiles of ceftiofur sodium and its related metabolites demonstrated their relatively slow, however, good absorption after subcutaneous administration, poor distribution, and slow elimination in cats. Based on the time of drug concentration above the minimum inhibitory concentration (MIC) (T>MIC) calculated in the current study, an intravenous or subcutaneous dose at 5 mg/kg BW of ceftiofur sodium once daily is predicted to be effective for treating feline bacteria with a MIC value of ≤4.0 μg/ml.

The pharmacokinetics of ceftiofur were studied in 78 catfish divided into 3 groups following IV and IM (single and repeated) administrations. Following a single IV injection of 5 mg/kg body weight of ceftiofur in normal catfish (Clarias lazera), serum concentration-time curve was best described by a two compartments open model with elimination half life (t0.5β), volume of distribution (Vdss) and total body clearance (CLtot) of 5.700 h, 229.71 ml/kg and 0.642 ml/kg/min, respectively. Following a single IM administration of 5 mg ceftiofur /kg body weight in normal catfish (Clarias lazera) , the peak serum concentration (Cmax) was 21.77 µg/ml, achieved at a maximum time (Tmax) of 2.15 h. The mean systemic bioavailability was 67.22%. The serum concentrations of ceftiofur following repeated IM administration of 5 mg/kg body weight once daily for five consecutive days in healthy and experimentally Aeromonas hydrophilia infected catfish (Clarias lazera) showed a lower significant values recorded in experimentally Aeromonas hydrophilia infected catfish (Clarias lazera) than in normal ones. Ceftiofur showed accumulative behavior in serum of fish. Results of this study indicated that ceftiofur was useful for treatment of Aeromonas hydrophilia infections in fish. Ceftiofur was assayed in serum, liver, kidney, dorsal muscle, abdominal muscle and skin after 24, 48, 72, 96 and 120 h from the last daily dose of 5mg ceftiofur/kg body weight for five days.. Ceftiofur could not be detected by spectrophotometer assay in all tested tissues in normal fish except in liver (four days), kidney (four days), and dorsal muscle (four days) post last administrations. Ceftiofur was completely cleared from serum and tissues at five days after the stoppage of drug dosage but in experimentally infected fish Ceftiofur could not be detected in all tested tissues except in liver (three days) and kidney (three days) post last administrations. Ceftiofur was completely cleared from all tissues at (four days) after the stoppage of drug dosage.

Despite the high number of antibiotics used for the treatment of infectious disease in animals, the development of slow release formulations presents a significant challenge, particularly in using novel biomaterials with low cost. In this report, we studied the pharmacokinetics, toxicity, and therapeutic activity of ceftiofur–PHBV (ceftiofur–poly(3-hydroxybutyrate-co-3-hydroxyvalerate)) in rats. The pharmacokinetic study demonstrated a sustained release of ceftiofur into the bloodstream, with detectable levels over the minimum inhibitory concentration for at least 17 days after a single intramuscular injection of ceftiofur–PHBV (10 mg/kg weight). In addition, the toxicological evaluation of biochemical, hematological, and coagulation blood parameters at the therapeutic dose demonstrated the safety of ceftiofur–PHBV, with no adverse effects. In addition, ceftiofur–PHBV exhibited a therapeutic effect for a longer time period than the nonencapsulated ceftiofur in rats challenged with Salmonella Typhimurium. The slow release of ceftiofur from the ceftiofur–PHBV, its low toxicity in the blood parameters evaluated, and the efficacy in the rats infected with Salmonella Typhimurium make ceftiofur–PHBV a strong candidate for biotechnological applications in the veterinary industry.
