Pharmacologic Inhibition of the TGF-β Type I Receptor Kinase Has Anabolic and Anti-Catabolic Effects on Bone

Abstract: During development, growth factors and hormones cooperate to establish the unique sizes, shapes and material properties of individual bones. Among these, TGF-β has been shown to developmentally regulate bone mass and bone matrix properties. However, the mechanisms that control postnatal skeletal integrity in a dynamic biological and mechanical environment are distinct from those that regulate bone development. In addition, despite advances in understanding the roles of TGF-β signaling in osteoblasts and osteoc… Show more

“…In addition, it impairs also adipogenesis, thus suggesting effects on a common progenitor cell [54]. TGFβ inhibition has potent anabolic effects [55] and restores osteogenesis in the presence of MM cellconditioned media and BM plasma from patients. It also suppresses MM cell growth.…”

“…Tumor-produced factors such as parathyroid hormone-related protein (PTHrP), interleukin-11 (IL-11), IL-6, and IL-8 stimulate bone resorption to release and activate growth factors housed in the mineralized bone matrix. Bone-derived TGF-b promotes osteolysis by stimulating tumor production of osteolytic factors, and alters the bone microenvironment to further increase osteoclastic bone resorption and inhibit bone formation Korpal et al 2009;Mohammad et al 2009). These tumor-bone interactions result in a feed-forward cycle to promote tumor growth in bone.…”

“…In multiple myeloma, TGF-β is secreted by myeloma and bone marrow stromal cells (19,20). TGF-β is an important factor during the disease development, which involves a complex signaling pathway network.…”
