The popularity of natural medicine is growing worldwide. Unlike conventional licensed medicines, herbal medicine practices are usually not supported by effectiveness, efficacy, or safety studies, which raise concerns about potential risks involved in their usage, particularly in high‐risk patients such as pregnant women where teratogenicity is a concern. Despite a lack of science‐based evidence, the use of herbal products for the management of pregnancy‐associated challenges is common, due to the common notion that they are free of toxic effects and adverse reactions because they are “natural.” The lack of concern about utilizing herbal remedies during pregnancy is strengthened by the lack of regulation in most countries for their marketing. However, plant‐based remedies are not free of adverse reactions. Medicinal plants and herbal remedies contain substances that can be toxic to the human body and the fetus. Potential effects of indiscriminate use of medicinal plants are embryotoxicity, teratogenic, and abortifacient effects. Some plant constituents can cross the placenta and reach the fetus. Phytochemicals and their metabolites are known to induce stimulation of uterine contraction and hormone imbalance that could result in abortion. The alterations to the hormonal profile can affect conception, induce teratogenic activity, and halt the pregnancy or produce a congenital malformation. Due to the wide range of modes of action of phytochemicals, some medicinal plants may be safe to use during certain trimesters of pregnancy and harmful at other stages. This manuscript reviews available scientific information concerning potential health hazards associated with the consumption of herbal medicines during pregnancy, highlighting those herbs that should be avoided due to their potential abortifacient and/or teratogenic activity. We focused on plants that were tested by preclinical studies, and studies of these plants are summarized. Common therapeutic use of these herbs, estimated effects, toxicological effects, and animal studies of these plants is summarized. The literature reviewed suggests that consumption of the following medicinal plants should be avoided during pregnancy: Abrus precatorius, Achyranthes aspera, Ailanthus excelsa, Aloe vera, Aristolochia indica, Areca catechu, Bambusa vulgaris, Cassia occidentalis, Cicer arietinum, Cimicifuga racemose, Dolichandrone falcate, Ginkgo biloba, Hydrastis canadensis, Indigofera trifoliate, Lavandula latifolia, Maytenus ilicifolia, Momordica cymbalaria, Moringa oleifera, Musa rosacea, Oxalis corniculate, Phytolacca dodecandra, Plumeria rubra, Ricinus communis, Ruta graveolens, Stachys lavandulifolia, Senna alata, Trigonella foenum‐graecum, Vitus agnus‐castus, and Valeriana officinalis.

Background: Triacontane and beta-sitosterol, though promising in various applications, face challenges due to low solubility and susceptibility to photosensitivity. Enhancing their effectiveness requires overcoming these limitations. Design of Experiment (DoE) coupled with High-Performance Thin Layer Chromatography (HPTLC) offers a potent approach to address these issues. Materials and Methods: The HPTLC method was optimized using a Box-Behnken Design (BBD), considering factors like saturation time, migration distance and application length. Response Surface Methodology (RSM) was employed to analyze critical risk factors' effects on the method. Validation parameters, including linearity, accuracy, precision, robustness, solution stability and forced degradation studies, were assessed according to ICH guidelines. Additionally, a different fractions of Ailanthus excelsa Roxb. plant extracts was analyzed. Results: Triacontane and beta-sitosterol displayed distinct R f values of 0.94±0.010 and 0.75±0.02 for triacontane and beta-sitosterol, respectively. The method demonstrated excellent linearity across the concentration range of 2-10 mg/band, with correlation coefficients of 0.999 triacontane and 0.9989 beta-sitosterol, respectively. Percentage recovery for both found within the range of 98.84-102.92%. This validated method offers specificity and robustness, ideal for routine quality control testing in pharmaceutical applications. Solution stability and forced degradation studies further supported the method's stability-indicating nature. Conclusion: The stability-indicating HPTLC method, developed and validated using DoE approach, proved suitable for simultaneous estimation of triacontane and beta-sitosterol in Ailanthus excelsa Roxb. plant extracts. Compliance with ICH guidelines, coupled with robustness and specificity, positions it as a valuable tool for quality evaluation and standardization of herbal formulations.

Effect of Isolated Allicin From Allium Sativum Linn Bulb for Effective Anti Fertility Activity in Female Wistar Rats

The present study was undertaken to evaluate antifertility activity of isolated allicin from garlic cloves and the raw juice of garlic (Allium sativum) (AS) on experimental female rats. Control of fertilization in female rats was investigated with estrogenic activity, evaluated using ethinyl estradiol as standard in immature female rats. Anti-implantation and early abortifacient activity was also investigated in female Wistar rats with the number of implantations and implantation resorptions estimations. Acute toxicity studies were carried out for the extracts and the test drug doses for screening were selected depending upon LD50 values. Fresh AS juice as well as isolated allicin (dose of 2.5 ml & 5.0 ml/kg b.w, and 10 mg/kg b.w respectively) was administered by oral route after confirmation of acute toxicity.In estrogenic activity evaluation, the juice of AS and isolated allicin (10 mg/kg p.o) showed significant estrogen-like activity at 5.0 ml/kg p.o. respectively by increasing the uterine weight compared to vehicle control group. Further the same treatment significantly decreased the number of implants and increased the number of resorptions compared to vehicle control group which were more significant for isolated compound rather extracts. Finally, the results revealed that the allicin isolated from AS ameliorates the scientific evidence of the antifertility activity as traditionally claimed.
