Allium mongolicum Regel (AM), widely distributed in western China, is a traditional Mongolian medicine herb. Two different solvents as water and methanol were used to extract AM, and their antioxidant capacity and inhibitory effects against key enzymes related to metabolic syndrome were assessed. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS and reducing power assays. In addition, the total phenolic content and total flavonoids content were quantificated and analyzed. Aqueous extract, having higher phenolic content (10.20 mg GAE/g DW) and flavonoid content (4.02 mg QE/g DW), showed better antioxidant and inhibitory effects against lipase and angiotensin‐converting enzyme (ACE); as for α‐glucosidase, the extract made by methanol showed better ability. In general, the aqueous extract of A. mongolicum Regel has the potential to be used as a functional food or nutraceutical in prevention and treatment of obesity and hypertension due to the high antioxidant and sound inhibitory potential against vital enzymes relevant to obesity and hypertension.

Bone marrow mesenchymal stem cell (BMSC) transplants are promising for the treatment of certain central nervous system diseases. However, oxidative stress is one of the major factors that may limit the survival of the transplanted BMSCs. The present study investigated the effect of pretreatment with gigantol on hydrogen peroxide (H2O2)‑induced apoptosis in rat BMSCs (rBMSCs) and the potential underlying mechanisms. The results demonstrated that gigantol pretreatment significantly inhibited H2O2‑induced apoptosis of rBMSCs. rBMSCs were incubated with 600 µM H2O2 in the absence or presence of different doses of gigantol (1‑100 µM). Cell viability and apoptosis ratios were assessed by MTT assays and flow cytometry, respectively. Morphological alterations and reactive oxygen species were measured by the fluorescent‑based methods of Hoechst staining and dichlorodihydrofluorescein diacetate, respectively. Furthermore, the protein levels of phosphorylated‑protein kinase B (Akt), B‑cell lymphoma‑2 (Bcl‑2), Bcl‑2‑associated X (Bax), caspase‑3 and caspase‑9 were investigated by western blotting. Following incubation with H2O2 for 2 h, gigantol significantly inhibited the H2O2‑induced reductions in the cell viability of rBMSCs in a dose‑dependent manner. Furthermore, gigantol upregulated Akt phosphorylation and Bcl‑2 expression, downregulated Bax expression, and reduced the expression of caspase‑3 and caspase‑9 in H2O2‑treated rBMSCs, whereas an opposite effect was detected when LY294002, an inhibitor of phosphatidylinositol 3‑kinase, was administered in combination with gigantol. These results indicate that gigantol may be developed as a promising neuroprotective agent for successful MSC transplantation in ischemic diseases.

Introduction:Active natural products from medicinal plants are good sources for the antioxidant and antidiabetic agent. Natural antioxidants protect human health against oxidative stress and free radicals which cause degenerative disease such as diabetes. Objectives: This study was conduct to try one approach to treat diabetes that is to reduce gastrointestinal glucose absorption through the inhibition of the α-glucosidase enzyme. Methods: We make three different root extracts from Durio zibethinus Murr. The extracts were examined for their DPPH radical scavenging activity, antioxidant activity using β-carotene-linoleic acid assay, and their α-glucosidase inhibitory activity by a standard method. Additionally, we have also determined their total polyphenols and total flavonoids quantitatively using spectrophotometer UV-Vis. Results: The root extracts Durio zibethinus Murr. were effectively scavenged DPPH radicals in varied rate. The hexane (EHD), ethyl acetate (EEAD), and ethanol (EED) extracts revealed DPPH radical scavenging activity with IC 50 of 541.28, 83.95 and 11.24 µg/ml respectively and their β-carotene-linoleic acid assay showed activity with IC 50 of 273.58, 139.53, and 166.83 µg/ml, respectively. In vitro assay of the α-glucosidase inhibitory activity of the EHD, EEAD, and EED extracts showed an IC 50 of 119.84, 23.69, and 3.35 µg/ml, respectively. Conclution: In this present study, we found that ethanol extract revealed the most active antioxidant activity and the highest inhibitory activity against α-glucosidase enzyme. The total phenolics and total flavonoids contents of the extracts were studied, where the ethanol extracts were found to have the highest than that of other extracts. This study proofs the medicinal potencies of Durio zibethinus Murr. root extracts.
